SLC15A4 promotes oral cancer progression by regulating TLR9 and activating the JAK/STAT signaling pathway

open access: yesHead & Face Medicine
Background Oral cancer continues to pose a major health problem worldwide, characterized by few treatment options and an unfavorable prognosis. The solute carrier family 15 member 4 (SLC15A4) has been reported to be associated with immune responses ...

TAB2 Causes Neuronal Damage by Aggravating Microglia‐Mediated Neuroinflammation in Parkinson's Disease

open access: yesAdvanced Science, EarlyView.
In microglia, STAT3 upregulates TAB2, which promotes NF‐κB activation through its NZF domain‐mediated recognition of K63‐linked ubiquitin chains, leading to inflammatory cytokine release and subsequent neuronal injury. Lumacaftor suppresses TAB2 expression and directly binds the TAB2‐NZF domain to interrupt K63 ubiquitin recognition, thereby blocking ...

TH17–Associated Polyamine Metabolism‐Guided Nanozyme Promotes Alveolar Bone Repair in Periodontitis

open access: yesAdvanced Science, EarlyView.
A pronounced TH17‐skewed immune response is identified in experimental periodontitis, and enrichment of the polyamine pathway is observed during TH17 differentiation, suggesting its potential as an immunometabolic target. Inspired by these findings, DFMO@Ui‐Mn is developed as a therapeutic platform combining cascade ROS scavenging with immunometabolic ...

The Evolution of IL6-IL6R-JAK-STAT Signaling Pathway in Metazoan

open access: yesBiology
Background: The Interleukin-6 (IL6)–Interleukin-6 receptor (IL6R)–Janus kinase (JAK)–signal transducer and activator of transcription (STAT) signaling pathway plays a crucial role in innate immunity by inducing the production of various immune effectors.

Low SLC8A1 in Colorectal Cancer Dictates Radiotherapy Resistance Through Calcium‐Dependent ANXA2+sEVs Driving M2 Polarization of TAMs

open access: yesAdvanced Science, EarlyView.
SLC8A1 deficiency in colorectal cancer cells increases intracellular calcium signaling, promoting the secretion of ANXA2‐enriched small extracellular vesicles. These vesicles reprogram tumor‐associated macrophages towards an M2 phenotype, which in turn fosters a tumor microenvironment conducive to radioresistance and is associated with poor patient ...

The Cancer Cell Metabolic Reprogramming Remodels the Tumor Microenvironment: Molecular Mechanisms and Therapeutic Strategies

open access: yesAdvanced Science, EarlyView.
This review elucidates how cancer cell metabolic reprogramming—across glucose, lipid, amino acid, and nucleotide pathways—remodels the tumor microenvironment to suppress anti‐tumor immunity and promote immune escape. Targeting these metabolic axes offers promising strategies to overcome immunotherapy resistance and enhance cancer treatment.

AARS1‐Mediated H3K27 Lactylation Rewires Glycolysis to Sustain Aggressive and Recurrent Bladder Cancer

open access: yesAdvanced Science, EarlyView.
AARS1 drives PI3K–AKT–mTOR‐dependent glycolysis in bladder cancer, promoting lactate accumulation, increased lactylation, H3K27la enrichment at the HK2 promoter, and HK2 transcription. HK2 reinforces glycolysis, sustaining a metabolic–epigenetic program associated with tumor progression, recurrence, and metastasis.

Spatiotemporal Multi‐Omic Mapping Reveals Liver‐Muscle Metabolic Crosstalk in Cancer Cachexia

open access: yesAdvanced Science, EarlyView.
The interactive CCAtlas platform delineates cross‐species, spatiotemporal, and sex‐specific molecular dynamics and metabolic rewiring across organs during cancer cachexia. Hepatic Gamt downregulation curtails hepatic creatine synthesis to trigger systemic creatine insufficiency and consequent skeletal muscle atrophy in LLC tumour‐bearing mice ...

JAK-STAT Pathway

open access: yes
Modulation of the antiviral innate immune response has been proposed as a putative cellular target for the development of novel pan-viral therapeutic strategies. The Janus kinase-signal transducer and activator of transcription (JAK-STAT) pathway is especially relevant due to its essential role in the regulation of local and systemic inflammation in ...

Sublattice‐Resolved Magnetoelastic Coupling and Opposing Pressure Responses in the Frustrated Magnetocaloric GdMn2O5

open access: yesAdvanced Science, EarlyView.
In GdMn2O5, sublattice‐resolved EXAFS and diffraction analysis reveal pronounced spin‐phonon coupling and an unexpected opposing pressure response: the Mn‐driven Néel temperature slightly decreases while the Gd‐sublattice ordering temperature rises under pressures up to 0.86 GPa.
