ATF4‐mediated stress response as a therapeutic vulnerability in chordoma

open access: yesMolecular Oncology, EarlyView.
We screened 5 chordoma cell lines against 100+ inhibitors of epigenetic and metabolic pathways and kinases and identified halofuginone, a tRNA synthetase inhibitor. Mechanistically halofuginone induces an integrated stress response, with eIF2alpha phosphorylation, activation of ATF4 and its target genes CHOP, ASNS, INHBE leading to cell death ...

Using deep networks for knee range of motion monitoring in total knee arthroplasty rehabilitation

open access: yesFrontiers in Bioengineering and Biotechnology
BackgroundKnee range of motion (ROM) is a key indicator of rehabilitation after total knee arthroplasty (TKA). Current tools, such as visual and protractor measurements, are cumbersome, imprecise, and require professional training, limiting their use in ...

Quantitative sensory testing in painful hand osteoarthritis demonstrates features of peripheral sensitisation. [PDF]

open access: yes, 2012
Hand osteoarthritis (HOA) is a prevalent condition for which treatments are based on analgesia and physical therapies. Our primary objective was to evaluate pain perception in participants with HOA by assessing the characteristics of nodal involvement ...

LDAcoop: Integrating non‐linear population dynamics into the analysis of clonogenic growth in vitro

open access: yesMolecular Oncology, EarlyView.
Limiting dilution assays (LDAs) quantify clonogenic growth by seeding serial dilutions of cells and scoring wells for colony formation. The fraction of negative wells is plotted against cells seeded and analyzed using the non‐linear modeling of LDAcoop.

Glaucocalyxin A delays the progression of OA by inhibiting NF-κB and MAPK signaling pathways

open access: yesJournal of Orthopaedic Surgery and Research
Background Osteoarthritis (OA) is a common degenerative joint condition marked by inflammation and cartilage breakdown. Currently, there is a dearth of treatment medications that can clearly slow the course of OA.

Plecstatin inhibits hepatocellular carcinoma tumorigenesis and invasion through cytolinker plectin

open access: yesMolecular Oncology, EarlyView.
The ruthenium‐based metallodrug plecstatin exerts its anticancer effect in hepatocellular carcinoma (HCC) primarily through selective targeting of plectin. By disrupting plectin‐mediated cytoskeletal organization, plecstatin inhibits anchorage‐dependent growth, cell polarization, and tumor cell dissemination.

Stress distribution of secondary bending in single-lap bolted joints with dissimilar joining plates and plate types [PDF]

open access: yes, 2016
Single-lap joints are an important class of bolted joint in the aerospace and civil engineering sectors. This type of joint is preferred as it can reduce weight and hence help to optimize fuel efficiency.
