Oxidative denaturation of Cu/Zn‐superoxide dismutase associated with neurodegenerative diseases

open access: yesProtein Science, Volume 34, Issue 11, November 2025.
Abstract Misfolding of mutant Cu/Zn‐superoxide dismutase (SOD1) is a well‐established pathological feature of familial amyotrophic lateral sclerosis (ALS). While amino acid substitutions in mutant SOD1 destabilize its structure and promote misfolding, oxidation has also been implicated in the pathological alterations of wild‐type SOD1, particularly in ...

Cytotoxicity of Atropisomeric [1,1′‐Binaphthalene]‐2,2′‐Diamines (BINAM) and Analogs in Human Cancer Cells: Enantioselectivity, Structure–Activity Relationships, and Mechanism

open access: yesChemMedChem, Volume 20, Issue 19, October 6, 2025.
It is reported for the first time that atropisomeric (R)‐[1,1′‐binaphthalene]‐2,2′‐diamine (R‐BINAM, 1(R)), but not (S)‐[1,1′‐binaphthalene]‐2,2′‐diamine (S‐BINAM, 1(S)), is a moderately potent spindle poison, causing antiproliferation, depolymerization of microtubules, multipolar spindles, pericentriolar material (PCM) fragmentation, mitotic ...

Loss of primary cilia promotes EphA2‐mediated endothelial‐to‐mesenchymal transition in the ovarian tumor microenvironment

open access: yesMolecular Oncology, Volume 19, Issue 10, Page 2951-2966, October 2025.
Loss of primary cilia in endothelial cells promotes EndMT and vascular abnormalities in the ovarian tumor microenvironment through EphA2 activation. Using human samples, in vitro models, and endothelial‐specific Kif3a‐knockout mice, we show that primary cilia loss drives the acquisition of cancer‐associated fibroblast‐like phenotypes, thereby ...

Intersection Between Local Anesthetics and Cancer Biology: What Now? Where Are We Going?

open access: yesAdvanced Biology, Volume 9, Issue 10, October 2025.
Local anesthetics (LAs), widely used in anesthesia, may also play a role in cancer treatment by inhibiting tumor growth, reducing metastasis, and enhancing immune responses. They modulate key pathways like AKT/mTOR and RAS/ERK while affecting angiogenesis and tumor innervation and the immune response.

Dynein‐Dependent Endo‐Lysosomal Degradation Drives Lewy Body Disorders Accompanied by Aβ Pathology

open access: yesAdvanced Science, Volume 12, Issue 37, October 6, 2025.
This study investigates the impact of Aβ plaques on α‐synuclein (αSyn) pathologies in dementia with Lewy bodies (DLB). Co‐culturing primary neurons with AD mouse brain slices reveals impaired dynein‐dependent organelle trafficking, affecting αSyn degradation. Activating Rab7 restores this process, reducing αSyn inclusions.
