Abstract Difelikefalin, a selective kappa opioid receptor agonist designed to limit central nervous system (CNS) penetration, is under development for the treatment of pruritus. Its hydrophilic, small‐peptidic structure limits CNS entry, minimizing potential CNS‐mediated adverse events (AEs).

In vitro evaluation suggests fenfluramine and norfenfluramine are unlikely to act as perpetrators of drug interactions. [PDF]
Mechanistic static pharmacokinetic models predict that CYP450 inhibition and induction are unlikely for fenfluramine or its major metabolite norfenfluramine at doses used for seizure reduction (0.2−0.7 mg/kg/day). These results suggest minimal drug‐drug interaction perpetrator potential for either fenfluramine or norfenfluramine.

Reaction phenotyping showed metabolism of the antiseizure medication fenfluramine by recombinant human cytochrome P450 (rCYP) enzymes rCYP2B6, rCYP1A2, rCYP2C19, and rCYP2D6. Fenfluramine dose may need to be adjusted when co‐administered with strong inhibitors or inducers of these enzymes (e.g., stiripentol).

Abstract Archaeological evidence as well as textual sources leave no doubt about Alwa's (Alodia's) intense transcultural connections, further corroborated by understudied overseas glass bead imports found there. This paper presents results of an analysis of 23 glass beads from Soba, the most prosperous capital of medieval Nubia.
