A light‐triggered Time‐Resolved X‐ray Solution Scattering (TR‐XSS) workflow with application to protein conformational dynamics

open access: yesFEBS Open Bio, EarlyView.
Time‐resolved X‐ray solution scattering captures how proteins change shape in real time under near‐native conditions. This article presents a practical workflow for light‐triggered TR‐XSS experiments, from data collection to structural refinement. Using a calcium‐transporting membrane protein as an example, the approach can be broadly applied to study ...

Generating clean few-cycle pulses in an all-bulk multipass cell scheme [PDF]

open access: yesEPJ Web of Conferences
We theoretically demonstrate the generation of clean few-cycle pulses in a three-stage all-bulk multipass cell scheme. By meticulously selecting the number of round trips and the width of the material used in each cell, we are able to keep the three ...

Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

Topological high-harmonic spectroscopy

open access: yesCommunications Physics
Linearly polarized vector beams are structured lasers whose topology is characterized by a well-defined Poincaré index, which is a topological invariant during high-order harmonic generation.

Self-calibrating d-scan: measuring ultrashort laser pulses on-target using an arbitrary pulse compressor

open access: yesScientific Reports, 2018
In most applications of ultrashort pulse lasers, temporal compressors are used to achieve a desired pulse duration in a target or sample, and precise temporal characterization is important.

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Análisis Geométrico y Microestructural de Recubrimientos de NiCoCrAlYTa proyectados por láser sobre láminas de acero inoxidable AISI 316L

open access: yes, 2014
[EN] In this work, the NiCoCrAlYTa Alloy was deposited by laser cladding with different processing parameters (flow rate of powder supplied, feed speed and laser power), in order to analyze the effects of processing on the geometry, microstructure and properties of the coating.

Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.
