Lac repressor — Lac operator complexes

European Biophysics Journal, 1987
Complexes between the Lac repressor and a small DNA operator fragment (29 base pairs) were investigated using polyacrylamide gel electrophoresis and solution X-ray scattering. Titration of the DNA fragment with the repressor, followed by gel electrophoresis showed that only two types of complexes are formed with repressor/operator ratios of 0.5 and 2 ...

How lac Repressor Binds to DNA

Nature, 1972
A sequence of about fifty amino-acids at the N-terminus of the lac repressor is thought to bind directly to lac operator DNA. Although this length cannot make the conventional cleft, it could form a protrusion; the manner in which such a protrusion interacts with the DNA site is discussed here.

Der lac‐Repressor

Angewandte Chemie, 1971

Operator search by mutant lac repressors

Journal of Molecular Biology, 1998
The Escherichia coli Lac and Gal repressors are two members of a large family of bacterial repressor proteins that share significant sequence and structural homology. Efficient repression by all family members requires specific binding to a site or sites close to the transcriptional start of the genes regulated.

Neutron scattering studies of lac repressor

Nature, 1980
The lac repressor, a tetrameric protein of identical subunits [molecular weight (MW) 4 x 38,500], interacts specifically with the lac operator, preventing the expression of the structural genes of the lac operon. The presence of an inducer (such as isopropyl-beta-D-thiogalactoside; IPTG) which binds to the repressor, prevents operator binding by ...

The Lac Repressor

1971
Publisher Summary This chapter discusses the lactose ( lac ) operon of Escherichia coli , its products, and its regulation. Jacob and Monod and their co-workers formulated their model of negative control of protein synthesis. This model has been confirmed by in vitro experiments, which permitted further analysis of the mechanism at the molecular ...

NMR Studies of lac Operator and lac Repressor

2003
For many years, and even now, the lac operon of Escherichia coli has been a model for gene regulation. The lac repressor is a tetrameric protein that binds strongly the lac operator and the affinity of which is modulated by an inducer molecule. The lac repressor–lac operator complex was the first nucleic acid–protein complex studied with success by NMR.

Genetic Analysis of the lac Repressor

1980
Understanding how proteins function is an essential part of molecular biology, since specific proteins or enzymes ultimately mediate all cellular processes. Unraveling the mysteries of prokaryotic and eukaryotic cells will require identification of the relevant proteins, and understanding how they work. Genetics is a powerful tool for the investigation

lac repressor-operator interaction. Analysis of the X86 repressor mutant

Journal of Molecular Biology, 1976
Abstract In vitro measurements show that the X86 repressor, which has an increased affinity for the lac operator as compared to wild-type repressor, also has an increased affinity for non-operator sites on Escherichia coli DNA. The rate constant of association of repressor and operator is decreased by E.
