Estimation and characterization of hamster leukocyte elastase

Abstract A method is described for the preparation of granule extract from hamster polymorphonuclear leukocytes. The presence of elastase in this extract has been ascertained by the dual criteria of elastin solubilization and synthetic elastase substrate hydrolysis. The extract is also able to cleave one out of three cathepsin-G substrates.

Polymorphonuclear leukocytes are known to accumulate in tissues subjected to ischemia and reperfusion. Studies on endothelial cell monolayers suggest that limited release of elastase plays an important role, via basement membrane degradation, in the leukocyte diapedesis and extravasation elicited by proinflammatory mediators.
