Membrane selectivity and pore formation of SprA1 and SprA2 hemolytic peptides from Staphylococcus aureus type I toxin–antitoxin systems

When overexpressed in Staphylococcus aureus, the type I toxins SprA1 and SprA2 form membrane pores, with SprA1 creating stable pores and SprA2 forming transient ones. Both induce concomitant membrane depolarization, ATP release, and growth arrest, while only SprA1 causes membrane permeabilization due to its stable pore formation.

A metastasis‐associated pannexin‐1 mutant (Panx11‐89) forms a minimalist ATP release channel

A truncation mutant of Panx1 (Panx11‐89), enriched in metastatic breast cancer cells, is capable of forming a large ATP‐permeable membrane channel despite containing only 20% of the amino‐terminal amino acids of the wild‐type channel protein (wtPanx1). However, cells expressing Panx11‐89 alone die, whereas co‐expression of wtPanx1 is protective.

Natural aminosterols inhibit NMDA receptors with low nanomolar potency

Misfunction of N‐methyl‐D‐aspartate (NMDA) receptors is associated with many brain disorders, making them primary targets for drug discovery. Trodusquemine and other natural aminosterols inhibit NMDA receptors with low nanomolar potency through binding to the cell membranes followed by interaction with the channels.

Extracellular vesicles in multiple myeloma: pathogenesis and therapeutic application
