Droplet networks, from lipid bilayers to synthetic tissues

open access: yes, 2019
An aqueous droplet in a solution of lipid in oil acquires a lipid monolayer coat. When two such droplets are brought together, they adhere through the formation of a droplet interface bilayer (DIB) (Fig. 1a).

Rab1A Promotes Hepatic Steatosis by Suppressing Mitophagy via the Raf‐1/ERK1/2/PINK1 Signaling Axis

open access: yesAdvanced Science, EarlyView.
Why does fat accumulate in the liver? Our study reveals Rab1A as the molecular switch that silences the cell's mitochondrial cleanup crew. Turning Rab1A off reactivates mitophagy, clears excess fat, and reverses fatty liver disease in mice—offering a promising new therapeutic target for MASLD, a growing global health challenge.

C18ORF32 modulates lipid droplet secretion via secretory autophagy and restrains hepatic steatosis in vivo

open access: yesCell Communication and Signaling
Background Lipid droplets (LDs) are essential for maintaining cellular homeostasis by storing excess lipids, but regulation of their cellular level is crucial to prevent lipotoxicity under normal conditions.

Mitochondria and Neuromast Tagging With Fluorescent Gallium‐Triapine Analogues: In Cellulo MP FLIM and Zebrafish Live Imaging

open access: yesAdvanced Science, EarlyView.
Fluorescent BODIPY‐conjugated thiosemicarbazone ligands and their Ga(III), In(III), and Fe(III) complexes, inspired by Triapine, are developed as theranostic agents. Multiphoton FLIM and confocal microscopy in cancer cells and zebrafish reveal real‐time uptake, mitochondrial localisation, and whilst spectroscopic assays indicated preserved complex ...

Opn3 Drives Blue-Light-Induced Reduction in Lipid Droplets and Antiviral Defense

open access: yesBiomolecules
Abnormal lipid metabolism is a key feature of many diseases. Therefore, investigating its underlying mechanisms is of great importance. Recently, blue light has shown promise as a drug-free way to influence energy metabolism, relying on the light ...

An Adipocyte MKK3–p38β Axis Preserves Insulin Responsiveness in Mice and Is Reciprocally Altered in Human Obesity

open access: yesAdvanced Science, EarlyView.
The adipocyte MKK3–p38β axis emerges as an adaptive stress‐response pathway during obesity. Human visceral adipose tissue analyses, adipocyte‐specific mouse models, and primary adipocytes show that the MKK3–p38β axis supports insulin responsiveness by restraining p70S6K‐dependent inhibitory IRS1 feedback and preserving AKT–GLUT4 signaling. This pathway

Lipid Droplet Surface Promotes 3D Morphological Evolution of Non‐Rhomboidal Cholesterol Crystals

open access: yesAdvanced Science
Cholesterol crystals, which cause inflammation and various diseases, predominantly grow in a platy, rhomboid structure on the plasma membranes but exhibit an uneven three‐dimensional (3D) architecture intracellularly.

Lipid-induced Caveolin1-Lipid droplet trafficking is associated with lipid droplet growth

open access: yes
Abstract Caveolae are 50-100 nm sized plasma membrane invaginations involved in cellular lipid uptake and lipid accumulation. Several caveolin1 and cavin1 mutations are associated to human lipodystrophy. Interestingly, previous research identified caveolae proteins at lipid droplets although their specific cellular function at lipid ...
