Combining LOPIT with differential ultracentrifugation for high-resolution spatial proteomics [PDF]

open access: yesNature Communications, 2019
Spatial proteomics allows studying cellular protein localisations at system-wide scale. Here, the authors show that combining the previously developed hyperLOPIT method with differential centrifugation yields protein localisation maps at suborganellar ...

Pattern Analysis of Organellar Maps for Interpretation of Proteomic Data [PDF]

open access: yesProteomes, 2022
Localization of organelle proteins by isotope tagging (LOPIT) maps are a coordinate-directed representation of proteome data that can aid in biological interpretation.

Conditional constructions in Lopit, an Eastern Nilotic language

open access: yesStudies in African Linguistics, 2017
Lopit is an Eastern Nilotic language of South Sudan. It has a number of ways of expressing conditionals. The most common way involves the use of the subordinate clause marker l- on the clause-initial verb which introduces the protasis.

CFTR Rescue by Lumacaftor (VX-809) Induces an Extensive Reorganization of Mitochondria in the Cystic Fibrosis Bronchial Epithelium [PDF]

open access: yesCells, 2022
Background: Cystic Fibrosis (CF) is a genetic disorder affecting around 1 in every 3000 newborns. In the most common mutation, F508del, the defective anion channel, CFTR, is prevented from reaching the plasma membrane (PM) by the quality check control of

Multilocation proteins in organelle communication: Based on protein–protein interactions

open access: yesPlant Direct, 2022
Protein–protein interaction (PPI) plays a crucial role in most biological processes, including signal transduction and cell apoptosis. Importantly, the knowledge of PPIs can be useful for identification of multimeric protein complexes and elucidation of ...

Spatial proteomics of vesicular trafficking: coupling mass spectrometry and imaging approaches in membrane biology. [PDF]

open access: yesPlant Biotechnol J, 2023
Summary In plants, membrane compartmentalization requires vesicle trafficking for communication among distinct organelles. Membrane proteins involved in vesicle trafficking are highly dynamic and can respond rapidly to changes in the environment and to cellular signals.

Spatiotemporal proteomic profiling of the pro-inflammatory response to lipopolysaccharide in the THP-1 human leukaemia cell line [PDF]

open access: yesNature Communications, 2021
“Protein relocalisation plays a major role in the innate immune response but remains incompletely characterised. Here, the authors combine temporal proteomics with LOPIT, a spatial proteomic workflow, in a fully Bayesian framework to elucidate ...

Localization of Organelle Proteins by Isotope Tagging (LOPIT) [PDF]

open access: yesMolecular and Cellular Proteomics, 2004
We describe a proteomics method for determining the subcellular localization of membrane proteins. Organelles are partially separated using centrifugation through self-generating density gradients. Proteins from each organelle co-fractionate and therefore exhibit similar distributions in the gradient.

Putative Glycosyltransferases and Other Plant Golgi Apparatus Proteins Are Revealed by LOPIT Proteomics   [PDF]

open access: yesPlant Physiology, 2012
AbstractThe Golgi apparatus is the central organelle in the secretory pathway and plays key roles in glycosylation, protein sorting, and secretion in plants. Enzymes involved in the biosynthesis of complex polysaccharides, glycoproteins, and glycolipids are located in this organelle, but the majority of them remain uncharacterized. Here, we studied the
