Interaction of HS1BP3 with cortactin modulates TKS5 localisation, cell secretion and cancer malignancy

open access: yesMolecular Oncology, EarlyView.
Here, we demonstrate that HS1BP3 interacts with Cortactin through a proline‐rich region (PRR3.1) and show that this interaction, and HS1BP3 itself, promote cancer cell proliferation and invasion. Inhibition of this interaction leads to build‐up of TKS5 in multivesicular endosomes and altered secretion of CD63 and CD9, providing an explanation for the ...

On lower bounds for the energy of graphs

open access: yesArab Journal of Basic and Applied Sciences
The concept of the energy of a graph was introduced by Gutman in 1978, inspired by the Hückel molecular orbital theory, which approximates the total [Formula: see text]-electron energy of a conjugated hydrocarbon molecule using the energy of its ...

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Tight analyses for subgradient descent I: Lower bounds

open access: yesOpen Journal of Mathematical Optimization
Consider the problem of minimizing functions that are Lipschitz and convex, but not necessarily differentiable. We construct a function from this class for which the $Tþ$ iterate of subgradient descent has error $\Omega (\log (T)/\sqrt{T})$. This matches

Developmental programmes drive cellular plasticity, disease progression and therapy resistance in lung adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
This study shows that lung adenocarcinomas exploit developmental branching morphogenesis to acquire a therapy resistant basal‐like tumour cell state. This process was found to be regulated by combined TP53 loss‐of‐function and type‐I interferon signalling, identifying a novel axis for biomarker and therapeutic target discovery.

Loss of IGF‐1R impairs DNA‐PKcs recruitment to chromatin leading to defective end‐joining

open access: yesMolecular Oncology, EarlyView.
IGF‐1R promotes radioresistance by facilitating DNA‐PKcs recruitment to chromatin, enabling non‐homologous end‐joining (NHEJ) repair of double‐strand breaks. Inhibition or loss of IGF‐1R disrupts this recruitment to damage sites, driving compensatory reliance on microhomology‐mediated end‐joining (MMEJ) repair.

Flow Enabled Target Capture Halbach‐based magnetic enrichment increases circulating tumor cell capture from blood in metastatic cancer patients

open access: yesMolecular Oncology, EarlyView.
Pair‐wise comparison of the CellSearch and FETCH enrichment technologies for circulating tumor cells (CTCs) from metastatic breast, prostate, and small cell lung cancer patients shows an increased capture of CTCs using FETCH enrichment. The clinical implementation of circulating tumor cells (CTCs) as a predictive tool for therapy efficacy in the ...

Approximate Lower Bound Arguments

open access: yes
Suppose a prover, in possession of a large body of valuable evidence, wants to quickly convince a verifier by presenting only a small portion of the evidence. We define an Approximate Lower Bound Argument, or ALBA, which allows the prover to do just that: to succinctly prove knowledge of a large number of elements satisfying a predicate (or, more ...

Lower bounds on the Haraux function

open access: yesJournal of Approximation Theory
The Haraux function is an important tool in monotone operator theory and its applications. One of its salient properties for a maximally monotone operator is to be valued in $[0,+\infty]$ and to vanish only on the graph of the operator. Sharper lower bounds for this function have been proposed in specific cases.
