Stereoselective Biotransformation: Transfer of Learning to Advance Drug Metabolism and Biocatalysis
Understanding stereoselective biotransformations has implications for predicting drug disposition and response and may also inspire novel biocatalytic and biomimetic strategies to address challenges in metabolite and API synthesis. ABSTRACT Chirality is an important determinant of drug action, as enantiomers can exhibit markedly different ...

As the major component in the cell wall of brown algae, alginates are degradable by alginate lyases via β-elimination. Alginate lyases can be categorized into various polysaccharide lyase (PL) families, and PL7 family alginate lyases are the largest ...

Alginate Lyases: Sources, Mechanism of Activity and Potencial Application
Alginate lyases are group of enzymes which catalyze depolymerization of alginate into oligosaccharides. Alginate lyase have been widely used in many applications such as in production of bioactive oligosaccharides, control of polysaccharide rheological ...

A dedicated haem lyase is required for the maturation of a novel bacterial cytochrome c with unconventional covalent haem binding [PDF]
In bacterial c-type cytochromes, the haem cofactor is covalently attached via two cysteine residues organized in a haem c-binding motif. Here, a novel octa-haem c protein, MccA, is described that contains only seven conventional haem c-binding motifs ...

Engineering a Virus‐Derived X Family DNA Polymerase FvPolX for de novo DNA Synthesis
In this study, a Faustovirus‐derived polymerase (FvPolX) is engineered based on structural and sequence analysis of PolX enzymes. The optimal mutants demonstrate substantially enhanced catalytic activity toward dNTPs and 3′‐ONH2‐dNTPs compared to the wild‐type enzyme.

Purification and characterization of alginate lyase from locally isolated marine Pseudomonas stutzeri MSEA04 [PDF]
An alginate lyase with high specific enzyme activity was purified from Pseudomonas stutzeri MSEA04, isolated from marine brown algae. The alginate lyase was purified by precipitation with ammonium sulphate, acetone and ethanol individually.

Dysregulated TCA cycle contributes to Alzheimer's disease (AD) pathogenesis. Here, we show that microglial isocitrate dehydrogenase 1 (IDH1) is a critical driver. Elevated IDH1 disrupts citrate metabolism and mitochondrial function, exacerbating AD pathology.

Discovery of a novel (R)-selective bacterial hydroxynitrile lyase from Acidobacterium capsulatum
Hydroxynitrile lyases (HNLs) are powerful carbon–carbon bond forming enzymes. The reverse of their natural reaction – the stereoselective addition of hydrogen cyanide (HCN) to carbonyls – yields chiral cyanohydrins, versatile building blocks for the ...
