In vitro testing of chlorpropamide with human lymphocyte cultures in the presence of liver microsome fraction S9 mix of rats.

The oral hypoglycaemic drug 1-(p-chloroben-zenesulfonyl)-3-propylurea (chlorpropamide) was examined for clastogenic properties using human lymphocytes with a microsomal (S9 mix) activation system. Lymphocytes, exposed to the S9 mix only as well as to the solvent DMSO, yielded chromosome aberration levels within the background range.

Prognostic value of HL-A typing and mixed lymphocyte culture tests in kidney transplantations from living related donors.

In 45 recipients of intrafamilial kidney transplants, the one year survival of sibling grafts was 95% and of parental grafts 71%. HL-A compatible grafts had a one year survival of 94% compared to 75% of HL-A incompatible grafts. At the same time, the survival of MLC compatible grafts was 100% and that of MLC incompatible grafts 74%.

A semi-micro method for the mixed lymphocyte culture (MLC) test in dogs is described. Optimization of various factors influencing the test were invistigated. Discrimination between allogeneic and isogeneic cell mixtures was possible after 90 h or culture but a culture period of 144 h was found to be optimal.

In comparison to the classical mixed lymphocyte culture (MLC) method a MLC technique using whole blood cultures of responding cells and a pool of prepared stimulating cells from 10 healthy blood donors is described. The reproducibility of cryopreservation of stimulating lymphocytes was proven by means of cell electrophoresis.

This study introduces mixed lymphocyte culture (MLC) as a clinical method for prognostic assessment in allogen grafts. The principle and technical data are discussed. The technical variance and biological reproducibility of the MLC was tested by means of the recommendations of the Sixth International Histocompatibility Workshop 1975.

Studies on the human mixed lymphocyte culture. II. Comparison of one-way and two-way test performed on lymphocytes of patients with end-stage renal failure and their prospective living donors.

The results of DNA synthesis in human one-way and two-way mixed lymphocyte cultures were compared. Twenty patients with chronic renal failure and their twenty six prospective living donors were investigated. Statistical correlation was found on the level alpha = 0.01 between the stimulation index value calculated for one-way and two-way technique.
