Machining characteristics of chromium-molybdenum steels: a case study of AISI 4140

open access: yesТехнічна інженерія
The article examines the machining characteristics of chromium-molybdenum steel AISI 4140, which is widely used in mechanical engineering due to its combination of high strength, wear resistance, and good machinability.

MITF maintains genome stability in nonmelanocyte lineages

open access: yesMolecular Oncology, EarlyView.
MITF is essential for melanocyte survival and acts as an oncogene in 10%–20% of melanomas. We show that MITF depletion causes genome instability in nonmelanocytic cells, leading to LATS2‐mediated P53 activation, cell cycle arrest, and apoptosis. This study highlights the role of MITF as a genome maintenance factor beyond the melanocyte lineage. Created

The effect of forging conditions and heat treatment on the machinability of titanium alloy workpieces

open access: yesТехнічна інженерія
In modern aircraft engine manufacturing, titanium alloys are an indispensable material for the production of the most critical and highly loaded rotor components of gas turbine engines, such as integrally bladed rotors (blisks) and compressor discs.

Liquid biopsy‐based diagnostic evaluation of hypermethylated CpG sites for ovarian cancer diagnosis

open access: yesMolecular Oncology, EarlyView.
This schematic outlines the workflow from biomarker identification to duplex MethyLight assay validation for epithelial ovarian cancer diagnosis using cfDNA‐based liquid biopsy. Initial screening of hypermethylated CpG candidates (cg02957270, cg10061138 cg00480298, COL2A1) was performed in tissue using ARMS‐PCR, COBRA, qPCR and image analysis. Selected

The machinability of nickel-based alloys in high-pressure jet assisted (HPJA) turning

open access: yesMetalurgija, 2013
Due to their mechanical, thermal and chemical properties, nickel-based alloys are generally included among materials that are hard to machine. An experimental study has been performed to investigate the capabilities of conventional and high-pressure jet ...

Patient therapy outcome modeling in cancer organoids is improved by cancer‐associated fibroblasts and organoid assembly convolution

open access: yesMolecular Oncology, EarlyView.
Patient‐derived organoids (PDOs) from pancreatic, colorectal, and gastric cancers were used to evaluate standard and experimental therapies. Incorporating cancer‐associated fibroblasts (CAFs) into organoid cultures improved patient therapy outcome prediction.

Epigenetic heterogeneity and plasticity in therapy‐induced tumor states through single‐cell multi‐omics

open access: yesMolecular Oncology, EarlyView.
Single‐cell multi‐omics reveals epigenetic heterogeneity across therapy‐adaptive tumor states, including quiescent/dormant, drug‐tolerant persister, and EMT‐like phenotypes. By linking regulatory features with state‐associated biomarkers, these approaches inform biomarker‐guided therapeutic strategies for evolving tumors.

In vitro and in silico modelling of ROS1‐positive non‐small cell lung cancer reveals fusion‐dependent tyrosine kinase inhibitor responses

open access: yesMolecular Oncology, EarlyView.
Drug resistance limits treatment success in a subset of lung cancers driven by ROS1 gene alterations. Using patient‐derived cells and computer simulations, we studied three key mutations and how they affect five targeted drugs. The mutations reduced drug effectiveness in different ways by altering protein structure and behavior.

Research and modelling of the high-speed milling process of heat-resistant high-alloy steel

open access: yesFrontiers in Mechanical Engineering
This study investigates the high‐speed milling behaviour of heat‐resistant martensitic–ferritic steel 15Kh12VMF, widely used in energy and power engineering components but difficult to machine due to its high hardness, strength and low thermal ...

Mycobacterial cell division arrest and smooth‐to‐rough envelope transition using CRISPRi‐mediated genetic repression systems

open access: yesFEBS Open Bio, EarlyView.
CRISPRI‐mediated gene silencing and phenotypic exploration in nontuberculous mycobacteria. In this Research Protocol, we describe approaches to control, monitor, and quantitatively assess CRISPRI‐mediated gene silencing in M. smegmatis and M. abscessus model organisms.
