Structural Basis of Specific Glucoimidazole and Mannoimidazole Binding by Os3BGlu7
β-Glucosidases and β-mannosidases hydrolyze substrates that differ only in the epimer of the nonreducing terminal sugar moiety, but most such enzymes show a strong preference for one activity or the other.

Theory and computation show that Asp463 is the catalytic proton donor in human endoplasmic reticulum α-(1→2)-mannosidase I [PDF]
It has been difficult to identify the proton donor and nucleophilic assistant/base of endoplasmic reticulum α-(1→2)-mannosidase I, a member of glycoside hydrolase Family 47, which cleaves the glycosidic bond between two α-(1→2)-linked mannosyl residues ...

TcALS knockdown accelerates metamorphosis via insulin signaling in Tribolium castaneum
RNAi silencing of TcALS accelerates larva‐to‐pupa metamorphosis in Tribolium castaneum. Abstract Insect metamorphosis is an evolutionary adaptation precisely regulated by nutritional and hormonal cues. The red flour beetle Tribolium castaneum, a major stored‐product pest, serves as an excellent model for studying this process.

Purification and properties of arylmannosidases from mung bean seedlings and soybean cells [PDF]
Two arylmannosidases (signified as A and B) were purified to homogeneity from soluble and microsomal fractions of mung bean seedlings. Arylmannosidase A from the microsomes appeared the same on native gels and on SDS gels as soluble arylmannosidase A ...

Glycosylation pattern of brush border-associated glycoproteins in enterocyte-like cells: involvement of complex-type N-glycans in apical trafficking [PDF]
We have previously reported that galectin-4, a tandem repeat-type galectin, regulates the raft-dependent delivery of glycoproteins to the apical brush border membrane of enterocyte-like HT-29 cells.

We identify a female patient with a homozygous nonsense variant (p.Gln38Ter) in the LYSET gene. This is the first western report of a challenging case of an extensive diagnostic odyssey and demonstrates that the LYSET gene must be considered in the differential diagnosis when M6P‐labeled lysosomal enzymes are altered.

A spectrophotometric assay for α-mannosidase activity [PDF]
A simple and versatile spectrophotometric assay for alpha-mannosidase activity, which can be used with unlabelled natural substrate or synthetic substrates, was developed. The reducing mannose released from the substrate by the enzyme is quantitated using glucose oxidase, peroxidase and o-dianisidine.

Tyrosinase degradation is prevented when EDEM1 lacks the intrinsically disordered region.
EDEM1 is a mannosidase-like protein that recruits misfolded glycoproteins from the calnexin/calreticulin folding cycle to downstream endoplasmic reticulum associated degradation (ERAD) pathway.

Xylan, the second most abundant plant cell wall polysaccharide, is degraded by β‐xylanases and β‐xylosidases. Here, we present the 2.65 Å cryo‐EM structure of Enterobacter cloacae β‐xylosidase (EcXyl43, GH43) and the 2.4 Å X‐ray structure of its inactive F507A mutant.
