Modeling Obesity and Weight Loss in Mice Using Novel AAV Approaches
ABSTRACT Objective This study utilized AAV gene delivery as an approach to induce and reverse hyperphagia in mice. We hypothesized that the delivery of orexigenic neuropeptides to the brain via AAV precipitates obesity and that the implementation of genetic switches to reverse transgene expression would elicit weight loss.

Bariatric surgery in a patient with complete MC4R deficiency [PDF]
Bariatric surgery is often successful for treatment of severe obesity. The mechanisms of weight loss after bariatric surgery and the role of central energy homeostatic pathways in this weight loss process are not well understood. The study of individuals with complete loss of function of genes important in the leptin-melanocortin system may help ...

AEGC conferred substantial resistance to weight gain and reduced food intake. AEGC may alleviate LR by regulating the levels of LEP and ADP. AEGC elevated the diversity of gut microbiota and reconstructed the composition. Changes such as CAG‐873 in gut microbiota may be the reason why AEGC alleviates LR.

Melanocortin Pathways: Suppressed and Stimulated Melanocortin-4 Receptor (MC4R)
Leptin-melanocortin pathway plays an essential role in the body weight regulation. Enhanced melanocortin signaling in the hypothalamus results in both decreased food intake and increased energy expenditure. The discovery of monogenic obesities with dysfunction of melanocortin-4 receptor (MC4R) greatly contributed to understanding of energy balance ...

Mitigation of Metabolic Diseases Through Personalized Nutrition: A Critical In‐Depth Review
The main objective of this review is to explore the impact of variation within the microbiome and among genes on nutrient metabolism and argue in favor of successful evidence of individualized dietary intervention for cardiovascular disease, diabetes, and obesity.

MRAP2 regulates energy homeostasis by promoting primary cilia localization of MC4R
The G protein–coupled receptor melanocortin-4 receptor (MC4R) and its associated protein melanocortin receptor–associated protein 2 (MRAP2) are essential for the regulation of food intake and body weight in humans.

Abstract Aims Acquired hypothalamic obesity (aHO) following craniopharyngioma (CP) is a challenging condition with limited therapeutic options. This study aimed to assess the real‐world effectiveness and safety of GLP‐1 RAs in this population. Materials and Methods CranioGLP1 is a retrospective multicentre study including 116 adults with aHO treated ...

Abstract Acquired hypothalamic obesity (aHO) is characterized by rapid and persistent weight gain resulting from structural or functional damage to the hypothalamus, typically accompanied by neuroendocrine dysfunction. While aHO is well described in the context of hypothalamic or suprasellar tumors, particularly craniopharyngioma, little is known about
