Model‐Driven Optimization of Subcutaneous Polymer Prodrugs Achieves Cancer Remission in Mice

open access: yesAdvanced Healthcare Materials, EarlyView.
A pharmacokinetics/pharmacodynamics (PK/PD) model was developed to evaluate multiple dosing regimens for subcutaneously administered water‐soluble polymer prodrug for cancer therapy. The model enabled prediction of in vivo performance and contributed to the optimization of anticancer efficacy.

Involvement of Cox-2 in the metastatic potential of chemotherapy-resistant breast cancer cells

open access: yesBMC Cancer, 2011
Background A major problem with the use of current chemotherapy regimens for several cancers, including breast cancer, is development of intrinsic or acquired drug resistance, which results in disease recurrence and metastasis.

Transcriptomic Changes in Cisplatin-Resistant MCF-7 Cells

open access: yesInternational Journal of Molecular Sciences
Breast cancer is a leading cause of cancer-related deaths among women. Cisplatin is used for treatment, but the development of resistance in cancer cells is a significant concern. This study aimed to investigate changes in the transcriptomes of cisplatin-resistant MCF7 cells.

Microengineered Gradient Hydrogels for Mechanobiology

open access: yesAdvanced Healthcare Materials, EarlyView.
Gradient hydrogels are used to mimic the mechanical heterogeneity in native tissues, offering powerful in vitro platforms to study cell‐material interactions in diverse pathophysiological contexts. Here, we present a comprehensive review of the design and experimental considerations for stiffness gradient hydrogels, discussing exemplary achievements ...

Human Umbilical Cord Mesenchymal Stem Cells Promote Breast Cancer Metastasis by Interleukin-8- and Interleukin-6-Dependent Induction of CD44/CD24 Cells

open access: yesCell Transplantation, 2015
Although emerging evidence links mesenchymal stem cells (MSCs) with cancer metastasis, the underlying mechanisms are poorly understood. In the present study, we found that human umbilical cord-derived MSCs (UC-MSCs) promoted MCF-7 cell migration in vitro

Anti-apoptotic action of zearalenone in MCF-7 cells

open access: yesEcotoxicology and Environmental Safety, 2005
Zearalenone (ZEA), a nonsteroidal estrogenic mycotoxin, is present in high concentrations in dairy products and cereals. Studies have indicated that ZEA could strongly provoke proliferation in estrogen-dependent breast cancer MCF-7 cells following estrogen ablation.

Hierarchically Engineered Multi‐Enzyme Nanoreactors for in vitro Drug Biosynthesis and Pathway Transplantation Into Cells

open access: yesAdvanced Materials, EarlyView.
A hierarchically structured metal–organic framework (eMIL) is shown to host the complete six‐enzyme biosynthesis pathway for the anti‐cancer pigment violacein, reshaping reaction dynamics and multiplying in vitro yields. eMIL nanoreactors deliver this reconstituted bacterial multi‐protein system into mammalian cells, where cell‐derived substrates and ...

Reversal Effect of Apatinib on P-gp-mediated Multidrug Resistance of Human Breast Cancer and Its Mechanisms

open access: yesZhongliu Fangzhi Yanjiu, 2018
Objective To investigate the reversal effect of apatinib on the multidrug resistance of human breast cancer and the underlying mechanisms. Methods MCF-7 and MCF-7/ADR cells were treated with different concentrations of apatinib for 48h.

An estrogen-dependent esterase activity in MCF-7 cells

open access: yesJournal of Steroid Biochemistry, 1987
The presence of steroidal esters in hormonally sensitive tissues lends importance to the esterases which convert the biologically inactive adducts to the parent potent forms. Accordingly, esterase-activities were studied in a human breast cancer model--the MCF-7 cell line. Tritiated estradiol esters- estradiol-17-acetate (EA), estradiol-17-valerate (EV)
