Methoxyphenyl Imidazolines as Potential Activators of p53

open access: yesMedical Sciences Forum, 2022
The use of p53-MDM2/X inhibitors is a prospective strategy in anti-cancer therapy for tumors with wild type p53 protein. In our study, new low-molecular-weight inhibitors of the p53-mdm2 interaction have been proposed.

Proteomic investigation of the MDM2 interactome and linear motif interactions [PDF]

open access: yes, 2011
The oncoprotein MDM2 has an integral role in cancer development via multiple signalling pathways. Two proteomic mass spectrometry screens, label-free with spectral counting quantitation and 8-plex iTRAQ were used to identify proteins up or downregulated ...

Retroperitoneal pararenal dedifferentiated liposarcoma with epithelioid/neuroendocrine features, somatostatin-positive.

open access: yesForum of Clinical Oncology, 2023
Liposarcomas are malignant mesenchymal neoplasms that, although derived from adipocytes, may dedifferentiate to other cell types. We present an exceedingly rare case of epithelioid/neuroendocrine dedifferentiation with somatostatin expression of a ...

CircHIPK3 Affects Malignant Phenotypes in Esophageal Squamous Cell Carcinoma by Regulating p53-Akt-Mdm2 Signaling Pathways

open access: yesZhongliu Fangzhi Yanjiu, 2019
Objective To investigate CircHIPK3 expression in esophageal squamous cell carcinoma (ESCC) tissues, so as to find a new way of diagnosis and targeted therapy. Methods qRT-PCR, CCK-8, Transwell and flow cytometry were performed to detect the difference of

Association between ТР53, MDM2 and NQO1 gene polymorphisms and viral load among women with human papillomavirus

open access: yesВавиловский журнал генетики и селекции, 2022
The risk of cervical cancer is caused by persistent human papillomavirus (HPV) infection. Cervical cancer is the most frequent cancer among women. Our purpose was to investigate the association between TP53 215C>G (Pro72Arg), MDM2 -410T>G, and NQO1

Understanding the Effect of Natural Products on Breast Cancer via P53-MDM 2 Signal Pathway [PDF]

open access: yesJournal of Cell and Molecular Research, 2021
The use of medicinal plants in the treatment of diseases has a long history dating back to the presence of humans on earth. Cancer has almost been an incurable disease, and among the various cancers, breast cancer is the most common type of cancer among ...

The mechanisms of regulation of Hdm2 protein level by serum growth factors [PDF]

open access: yes, 2006
Cell cycle progression in response to serum growth factors is dependent on the expression of functional Hdm2 (Mdm2), which inhibits p53-dependent transcription of anti-proliferative genes.

The MDM2 gene family [PDF]

open access: yesBioMolecular Concepts, 2013
Abstract MDM2 is an oncoprotein that blocks p53 tumor suppressor-mediated transcriptional transactivation, escorts p53 from the cell nucleus to the cytoplasm, and polyubiquitylates p53. Polyubiquitylated p53 is rapidly degraded in the cytoplasm by the 26S proteasome. MDM2 is abnormally upregulated in several types

Bivalent binding of p14ARF to MDM2 RING and acidic domains inhibits E3 ligase function

open access: yesLife Science Alliance, 2022
This study shows that MDM2 acidic and zinc finger regions weakly interact with the RING domain and a fragment of p14ARF strengthens this interaction to inhibit MDM2 ubiquitin ligase activity.

p300/MDM2 Complexes Participate in MDM2-Mediated p53 Degradation [PDF]

open access: yesMolecular Cell, 1998
Control of p53 turnover is critical to p53 function. E1A binding to p300/CBP translates into enhanced p53 stability, implying that these coactivator proteins normally operate in p53 turnover control. In this regard, the p300 C/H1 region serves as a specific in vivo binding site for both p53 and MDM2, a naturally occurring p53 destabilizer.
