This study identifies the mechanosensor PIEZO1 as a key factor in promoting acute myeloid leukemia progression and uncovers a ferroptosis defense system mediated by PIEZO1‐HIF1A‐SLC7A11 axis that is essential for leukemia stem cell maintenance. This ferroptotic defense system may represent a unique vulnerability for leukemia stem cells, providing a ...

Exploring the interplay between Piezo1 and YAP/TAZ in mechanotransduction
reservedMechanical signals are essential factors for cells profoundly impacting their phenotype. Mechanotransduction is the ability of cells to perceive physical and mechanical stimuli from the environment and transduce them into a biological response ...

An intracellular magneto‐mechanical platform utilizing MSC‐targeted nanomotors is developed. After intra‐articular delivery, these nanomotors target endogenous BMSCs and are actuated within lysosomes to execute trans‐planar rotational‐bouncing motions under a rotating‐fluctuating 3D magnetic field, generating amplified mechanical stimulation.

Full androgen deprivation (FAD) induces paracrine cholesterol in prostate cancer that drives the polarization of cancer‐associated fibroblasts (CAFs) and subsequent elevated matrix stiffness. Matrix stiffness in turn potentiates tumor cell survival under FAD pressure via dual mechanotransductive activation of the IRE1α‐XBP1s stress‐response axis ...

Converging and Unique Mechanisms of Mechanotransduction at Adhesion Sites
The molecular mechanisms by which physical forces control tissue development are beginning to be elucidated. Sites of adhesion between both cells and the extracellular environment [extracellular matrix (ECM) or neighboring cells] contain protein ...

FOS3D: A Fluorescence‐Enabled Toolkit for Characterizing a Three‐dimensional Osteosarcoma Model
FOS3D describes fluorescent (F) osteosarcoma (OS) cells in a tri‐dimensional (3D) model. The study comprises three phases: development, where biofabrication parameters are tuned to achieve cytocompatibility and tumor‐specific mechanical properties in cell‐laden gelatin methacryloyl constructs; validation, where whole‐well fluorescence reading is ...

Models of hair cell mechanotransduction
Hair cell mechanotransduction is based on a finely tuned machinery residing in the hair bundle, the hair cells receptive organelle. The machinery consists of a transduction channel, an adaptation motor, the tip link, and many other components that ...

Mechanical Activation of Piezo1 Drives Osteoarthritis Through Kdm5c‐Mediated Epigenetic Silencing
Excessive mechanical stress activates Piezo1, triggering Ca2+‐dependent cytoskeletal forces that deform the nucleus and reduce H3K4me3. Kdm5c demethylates H3K4me3 at Col2a1 and Runx3 promoters. Kdm5c knockout rescues degradation. Repurposed telmisartan directly inhibits Kdm5c, blocking this axis and showing disease‐modifying efficacy in mouse OA models

Supramolecular hydrogels enable real‐time visualization of mechanical stress through supramolecular switching mechanotransduction. Adamantane‐modified cellulose functions as a responsive domain that undergoes an on–off transition between hydrated and dehydrated states via reversible β‐cyclodextrin–adamantane complexation and decomplexation.
