Pharmacokinetics of ethinyl estradiol and mestranol

Pharmacokinetally, a 50 micrograms oral dose of mestranol (which itself is inactive) is bioequivalent to a 35 micrograms dose of ethinyl estradiol. Physiologically, mestranol ranges from 50% to 100% of the activity of ethinyl estradiol, depending on the endpoint chosen.

NMR Analysis of Mestranol Bulk Drug

A double assay procedure for mestranol is described. The method is based upon measurement of the NMR spectrum of mestranol in pyridine, using diphenylacetic acid as an internal standard. The signals chosen are those from the methoxyl and ethinyl groups. Three commercial lots of the steroid were studied, and a TLC study of each lot is described.

THE AVAILABILITY of a 19-nor progestational compound mixed with a small amount of estrogenic substance provided the opportunity to study the effect of the pseudopregnant state of the course of rheumatoid arthritis in a selected group of patients. 1. Following a single intravenous administration of [6,7–3H]ethynyloestradiol and [6,7–3H]mestranol to female rats, 55% of the labelling was excreted in the faeces within 3 weeks, whereas only small amounts of radioactivity (1.5%) were recovered from urine during the first 3 days.

ABSTRACT The present concept is that mestranol must be demethylated to ethinyl-oestradiol in order to have oestrogenic activity. This activation of mestranol has been studied in vivo by means of radiospirometry and in vitro incubations with rat liver microsomes.
