Treatment of lumi-estrone 3-methyl ether (I) with acetylene gave the C-17-epimeric compounds lumi-mestranol (3-methoxy-17 alpha-ethynyl-13 alpha-estra-1,3,5(10)-trien-17 beta-ol, III ) and epi-lumi-mestranol (3-methoxy-17 beta-ethynyl-13 alpha-estra-1,3,5(10)-trien-17 alpha-ol, IV).

Norethynodrel-Mestranol and Thyroid Function

Indices of thyroid function were measured in 10 euthyroid females after equilibration to each of 3 treatments: (a) without medication, (b) while taking 250 μg thyroxine daily, and (c) while taking 250 μg thyroxine and 10 mg norethynodrel-mestranol daily. The thyroidal radioiodine clearance rate decreased from treatment (a) to treatment (b).

Mestranol in Ovulation Suppression

The effect of ethinyl estradiol 3-methyl ether (EE3-ME) was investigated in 10 patients administered .10 mg. of Mestranol (EE3-ME) for 2 to 6 cycles. 100 mg progesterone was administered on Day 25 to control withdrawal bleeding. 100% Suppression of ovulation was demonstrated. Breakthrough bleeding occurred only in 1 patient during 1 cycle.

Effects of Norethynodrel With Mestranol on the Rabbit Eye

Ocular and systemic effects developed during and after administration of large doses of oral contraceptives in female rabbits. A slight pressure decrease in normal and hereditarily buphthalmic eyes was observed. Remarkable lowering of intraocular pressure was observed in an eye with fluctuating glaucoma secondary to a dislocated lens.

Norethynodrel with Mestranol in the Treatment of Acne

Norethynodrel with mestranol was administered for periods ranging from 3 through 24 cycles to 40 female patients for treatment of acne of varying degrees of severity. Abstract Metabolic effects of mestranol, given in continuous high dosage for 3 or more months, were investigated in 4 postmenopausal women with metastatic breast cancer. Three women improved subjectively; in 1 patient objective remission of disease occurred.
