Oxidation of methionine to methionine sulfoxide as a side reaction of cyanogen bromide cleavage

Analytical Biochemistry, 1982
Abstract In addition to cleaving the peptide bond in N-pentafluorobenzoyl-methionyl-glycine, cyanogen bromide can also oxidize this peptide to the corresponding sulfoxide, particularly at a higher pH. The yields of this sulfoxide are 1, 8, 64, and 97% when the reaction conditions are 70% formic acid, 0.1 m hydrochloric acid, pH 3.5 citrate buffer ...

Oxidation of Methionine Residues in Aqueous Solutions:  Free Methionine and Methionine in Granulocyte Colony-Stimulating Factor

Journal of the American Chemical Society, 2004
The free energy barriers and a mechanism of the oxidation of the amino acid methionine in water and in granulocyte colony-stimulating factor (G-CSF) are analyzed via combined quantum mechanical and molecular mechanical (QM/MM) methods, constrained molecular dynamics, and committor probability calculations.

Mediating Molecular Recognition by Methionine Oxidation:  Conformational Switching by Oxidation of Methionine in the Carboxyl-Terminal Domain of Calmodulin

Biochemistry, 2005
The C-terminus of calmodulin (CaM) functions as a sensor of oxidative stress, with oxidation of methionine 144 and 145 inducing a nonproductive association of the oxidized CaM with the plasma membrane Ca(2+)-ATPase (PMCA) and other target proteins to downregulate cellular metabolism.

The oxidative products of methionine as site and content biomarkers for peptide oxidation

Journal of Peptide Science, 2010
AbstractBiomarkers for peptide/protein oxidation under oxidative stress (OS) hold both incredible application potential as well as significant challenges. In this article, liquid chromatography and mass spectrometry were applied to establish a new method for evaluating the oxidation site and degree of peptide oxidized, with its oxidative product ...

The regulation of one-carbon oxidation in the rat by nitrous oxide and methionine

Archives of Biochemistry and Biophysics, 1982
Abstract Formate is oxidized to CO2 in the rat by folate-dependent reactions. Nitrous oxide treatment inhibited hepatic methionine synthetase activity, reduced hepatic S-adenosyl- l -methionine (Ado-Met) and tetrahydrofolate (H4 folate) concentrations and decreased the rate of formate oxidation in the rat.

Methionine oxidation as a major cause of the functional impairment of oxidized actin

Free Radical Biology and Medicine, 2002
A significant specific increase in the actin carbonyl content has been recently demonstrated in human brain regions severely affected by the Alzheimer's disease pathology, in postischemic isolated rat hearts, and in human intestinal cell monolayers following incubation with hypochlorous acid (HOCl). We have very recently shown that exposure of actin to

Stereospecific oxidation of methionine to methionine sulphoxide by tetrachloroauric(III) acid

Journal of the Chemical Society, Chemical Communications, 1973
The use of gold(III) as oxidant provides a method for the sterospecific oxidation of methionine to methionine sulphoxide under conditions that can be extended to biological systems containing sulphides; preliminary kinetic studies give a possible explanation of the sterospecifiction of this reaction.
