Cr(VI)-induced methylation and down-regulation of DNA repair genes and its association with markers of genetic damage in workers and 16HBE cells

Environmental Pollution, 2018
To examine the mechanism of hexavalent chromium [Cr(VI)]-induced carcinogenesis, a cross-sectional study in workers with or without exposure to Cr(VI) as well as in vitro administration of Cr(VI) in 16HBE cells was conducted. We explored the associations

Ultrasensitive DNA Methylation Ratio Detection Based on the Target-Induced Nanoparticle-Coupling and Site-Specific Base Oxidation Damage for Colorectal Cancer.

Analytical Chemistry, 2022
DNA methylation analysis holds great promise in the whole process management of cancer early screening, diagnosis, and prognosis monitoring. Nevertheless, accurate detection of target methylated DNA, especially its methylation ratio in the genome ...

Role of oxidative stress, genome damage and DNA methylation as determinants of pathological conditions in the newborn: an overview from conception to early neonatal stage.

Mutation research, 2020
Increasing evidence suggests that early-life events can predispose the newborn to a variety of health issues in later life. In adverse pre- and perinatal conditions, oxidative stress appears to play an important role in the development of future ...

Development of oxidation damage base-based fluorescent probe for direct detection of DNA methylation.

Analytical Chemistry, 2020
DNA methylation has become a promising epigenetic biomarker for cancer diagnosis, prognosis and therapy monitoring. Herein, we demonstrate for the first time the development of a new oxi-dation damage base (8-oxo-7,8-dihydroguanine (8-oxoG))-modified ...

The repair of DNA methylation damage in Saccharomyces cerevisiae

Current Genetics, 1996
The major genotoxicity of methyl methanesulfonate (MMS) is due to the production of a lethal 3-methyladenine (3MeA) lesion. An alkylation-specific base-excision repair pathway in yeast is initiated by a Mag1 3MeA DNA glycosylase that removes the damaged base, followed by an Apn1 apurinic/ apyrimidinic endonuclease that cleaves the DNA strand at the ...

Methylation damage response in hematopoietic progenitor cells

DNA Repair, 2007
The cellular response to methylation DNA damage was compared in multipotent CD34(+) hematopoietic stem cells and mature CD34(-) cells isolated from cord blood of the same donor. Cytofluorimetric analysis of freshly isolated cord blood cells indicated that both cell types were in the G0/G1 phase of the cell cycle.

Reactive oxygen species regulate miR‐17‐5p expression via DNA methylation in paraquat‐induced nerve cell damage

Environmental Toxicology, 2020
There is emerging evidence suggesting that oxidative stress and DNA methylation can alter miRNA expression. However, little is known on the mechanism of miR‐17‐5p expression changes in paraquat (PQ)‐induced nerve cell damage.

Transcriptome and DNA methylation changes modulated by sulforaphane induce cell cycle arrest, apoptosis, DNA damage, and suppression of proliferation in human liver cancer cells.

Food and Chemical Toxicology, 2019
Abnormal epigenetic alterations are one of the keystones of cancer development. Epigenetic targeting drugs have become a promising and effective cancer therapy strategy.

Repairing DNA-methylation damage

Nature Reviews Molecular Cell Biology, 2004
Methylating agents modify DNA at many different sites, thereby producing lethal and mutagenic lesions. To remove all the main harmful base lesions, at least three types of DNA-repair activities can be used, each of which involves a different reaction mechanism.
