Epigenetic Dysregulation of the NKX2‐1/SPDEF Axis Drives Persistent Goblet Cell Differentiation and Epithelial Barrier Dysfunction in Chronic Obstructive Pulmonary Disease

open access: yesRespirology, EarlyView.
This study investigated airway epithelial barrier dysfunction persistence in ex‐smokers with COPD and underlying mechanisms using single‐cell RNA and ATAC sequencing. Epigenetic NKX2‐1/SPDEF dysregulation maintains goblet cell hyperplasia and barrier dysfunction despite long‐term smoking cessation.

NQO1‐Responsive Prodrug for in Cellulo Release of Cytochalasin B as Cancer Cell‐Targeted Migrastatic

open access: yesSmall, EarlyView.
The design, synthesis, biochemical, and cell biological evaluation of the NQO1‐targeted prodrug BQTML‐CB is reported. BQTML‐CB selectively releases cytochalasin B upon activation by the quinone‐oxidoreductase NQO1 associated with invasiveness of cancer cells, representing a novel approach to improve cancer selectivity and reduce immunosuppressive ...

In Situ, Real‐Time Imaging of pH Gradients and Solution Flows through Silica Garden Membranes during Tube Growth

open access: yesSmall, EarlyView.
A fluorescent pH probe, added to a saturated metal ion solution, allows for mapping pH during the growth of a silica garden. The probe shows a very intense blue fluorescence at pH around 4 and an intense green fluorescence at pH above 6. The red component of the image allows detection of the formation of particles and tracking solution flows in the ...

A Simple, Ultrastable, and Cost‐Effective Oxygen‐Scavenging System for Long‐Term DNA‐PAINT Imaging

open access: yesSmall, EarlyView.
A simple, enzyme‐free, and cost‐effective oxygen scavenging system enhances DNA‐PAINT super‐resolution microscopy by increasing photostability, minimizing docking strand degradation, and enabling faster, high‐precision localizations. Its long‐term stability supports extended imaging, yielding improved spatial resolution and image quality.

Proteomic Profiling of Alveolar Macrophages Identifies Loss of Lysosomal Content as an Indicator of Nanofiber‐Induced Frustrated Phagocytosis

open access: yesSmall, EarlyView.
This proteomic study examines how nanofibers affect lung macrophages by analyzing protein changes following exposure to various fibrous materials. Nanofibers triggered the release of inflammatory enzymes, and importantly, lysosomal content release was identified as an early event associated with frustrated phagocytosis. Seventeen sensitive and relevant
