When spring ephemerals fail to meet pollinators : mechanism of phenological mismatch and its impact on plant reproduction [PDF]
The flowering phenology of early-blooming plants is largely determined by snowmelt timing in high-latitude and high-altitude ecosystems. When the synchrony of flowering and pollinator emergence is disturbed by climate change, seed production may be ...

Interpreting the effects of DNA polymerase variants at the structural level
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

A reversible histone H3 acetylation cooperates with mismatch repair and replicative polymerases in maintaining genome stability. [PDF]
Mutations are a major driving force of evolution and genetic disease. In eukaryotes, mutations are produced in the chromatin environment, but the impact of chromatin on mutagenesis is poorly understood.

One of the most common ways to assess ice-sheet reconstructions of the past is to evaluate how they impact changes in sea level through glacial isostatic adjustment. PaleoMIST 1.0, a preliminary reconstruction of topography and ice sheets during the past

Theoretical study on enhancement of DNA conductivity by copper modification of mismatched base pairs
Abstract DNA is a potential material for nanodevices. Metallization is a promising way to improve its conductivity. Two copper modified mismatched base pairs were designed by H-by-Cu replacement. The conductivities of these base pairs were studied by density functional theory (DFT) combined with nonequilibrium Green’s function (NEGF ...

Finding novel vulnerabilities of hypomorphic BRCA1 alleles
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

Many patients with urothelial cancer do not benefit from treatment with pembrolizumab, while at risk of severe side effects. Changes in the levels of circulating tumor DNA early during treatment, measured by a simple and affordable assay that can be easily implemented in the clinic, can be used as a prognostic tool to identify these patients.

The pro bono interests of law firm lawyers tend to differ from the actual legal needs of the poor. This difference results in the mismatch problem or the incongruence between the interests of firm lawyers and the needs of the poor.

Liquid biopsy‐based diagnostic evaluation of hypermethylated CpG sites for ovarian cancer diagnosis
This schematic outlines the workflow from biomarker identification to duplex MethyLight assay validation for epithelial ovarian cancer diagnosis using cfDNA‐based liquid biopsy. Initial screening of hypermethylated CpG candidates (cg02957270, cg10061138 cg00480298, COL2A1) was performed in tissue using ARMS‐PCR, COBRA, qPCR and image analysis. Selected

Detection of single base-pair mismatches in DNA by chemical modification followed by electrophoresis in 15% polyacrylamide gel. [PDF]
We have developed a method for distinguishing fragments of DNA that contain single-base mismatches from their perfectly paired homologues. Single-stranded regions within a duplex fragment are accessible to 1-cyclohexyl-3-(2-[4-(4-methyl)morpholinyl]ethyl)carbodiimide, which reacts with unpaired guanidylate and thymidylate residues in DNA. Intact linear
