Rapid degradation of mutant SLC25A46 by the ubiquitin-proteasome system results in MFN1/2-mediated hyperfusion of mitochondria. [PDF]

open access: yes, 2017
SCL25A46 is a mitochondrial carrier protein that surprisingly localizes to the outer membrane and is distantly related to Ugo1. Here we show that a subset of SLC25A46 interacts with mitochondrial dynamics components and the MICOS complex.

Targeting Lactate and Lactylation in Cancer Metabolism and Immunotherapy

open access: yesAdvanced Science, EarlyView.
Lactate, once deemed a metabolic waste, emerges as a central regulator of cancer progression. This review elucidates how lactate and its epigenetic derivative, protein lactylation, orchestrate tumor metabolism, immune suppression, and therapeutic resistance.

RIPK3 dampens mitochondrial bioenergetics and lipid droplet dynamics in metabolic liver disease

open access: yesHepatology, EarlyView., 2022
RIPK3 dampens mitochondrial bioenergetics and lipid droplet dynamics in metabolic liver disease. Abstract Background and Aims Receptor‐interacting protein kinase 3 (RIPK3) mediates NAFLD progression, but its metabolic function is unclear. Here, we aimed to investigate the role of RIPK3 in modulating mitochondria function, coupled with lipid droplet (LD)

Targeting MCL1 by UMI-77 to induce mitophagy alleviates renal tubular epithelial cells injury in acute kidney injury

open access: yesScientific Reports
Acute kidney injury (AKI) is a common clinical disease with complex pathophysiological processes and with high morbidity and mortality. Severe mitochondrial damages, such as mitochondrial swelling and fragmentation, disruption of membrane integrity, and ...

Human cachexia induces changes in mitochondria, autophagy and apoptosis in the skeletal muscle [PDF]

open access: yes, 2019
Cachexia is a wasting syndrome characterized by the continuous loss of skeletal muscle mass due to imbalance between protein synthesis and degradation, which is related with poor prognosis and compromised quality of life.

Time‐Controlled Refrigerated Stem Cell Therapy Mitigates Scleroderma Fibrosis via Modulation of Mitochondrial Autophagy and Gut Metabolism

open access: yesAdvanced Science, EarlyView.
This study established an RT‐MSCs‐based therapeutic approach for scleroderma in mice. RT‐MSCs attenuated fibrosis by regulating mitochondrial autophagy and restored gut microbiota homeostasis. Metabolomic analyses confirmed recovery of key metabolites, and RT‐MSCs demonstrated favorable lesion targeting and safety profiles.

An AI-guided screen identifies probucol as an enhancer of mitophagy through modulation of lipid droplets

open access: yesPLoS Biology, 2023
Failures in mitophagy, a process by which damaged mitochondria are cleared, results in neurodegeneration, while enhancing mitophagy promotes the survival of dopaminergic neurons.

Autophagy: A cyto-protective mechanism which prevents primary human hepatocyte apoptosis during oxidative stress [PDF]

open access: yes, 2012
The role of autophagy in the response of human hepatocytes to oxidative stress remains unknown. Understanding this process may have important implications for the understanding of basic liver epithelial cell biology and the responses of hepatocytes ...

GPCRs in CAR‐T Cell Immunotherapy: Expanding the Target Landscape and Enhancing Therapeutic Efficacy

open access: yesAdvanced Science, EarlyView.
Chimeric antigen receptor T cell therapy faces dual challenges of target scarcity and an immunosuppressive microenvironment in solid tumors. This review highlights how G protein‐coupled receptors can serve as both novel targets to expand the therapeutic scope and functional modules to enhance CAR‐T cell efficacy.

Unlocking immune responses via mitophagy genes: A dual-pronged strategy against hematologic tumors

open access: yesLabMed Discovery
Mitophagy plays a complex role in cancer biology, particularly in hematologic malignancies. It can promote tumor survival and drug resistance by removing damaged mitochondria and reducing mitochondrial reactive oxygen species (ROS) or conversely inhibit ...
