Covalent Organic Frameworks‐Delivered Reuterin Drives Trained Immunity in Tumor‐Associated Macrophages to Enhance Melanoma Immunotherapy via Glycerophospholipid Metabolism

Covalent organic framework (COF)‐reuterin induces trained immunity in tumor‐associated macrophages, promoting the generation of nitric oxide and reactive oxygen species, thereby enhancing the antitumor immune response. Additionally, COF‐Reuterin directly kills tumor cells while simultaneously eradicating pathogenic intratumoural bacteria.

ZDHHC12 Palmitoylates HDAC8 to Promote the Progression of Hepatocellular Carcinoma Associated with a Diet High in Saturated Fatty Acids

This study identifies ZDHHC12 as a critical mediator of PA‐induced HCC progression. Mechanistically, PA upregulates ZDHHC12 expression through SMARCA4 activation, leading to HDAC8 palmitoylation at Cys244 which blocks HSC70‐mediated lysosomal degradation of HDAC8, thereby promoting HCC development. FBXO2 Alleviates Intervertebral Disc Degeneration via Dual Mechanisms: Activating PINK1‐Parkin Mitophagy and Ubiquitinating LCN2 to Suppress Ferroptosis

F‐box only protein 2 (FBXO2) combats intervertebral disc degeneration (IVDD) by coordinating dual protection. It activates PTEN‐induced putative kinase 1 (PINK1)‐Parkin mitophagy restoring mitochondrial function and targets lipocalin‐2 (LCN2) for degradation, suppressing ferroptosis.
