Pitfalls and other issues with the MTT assay

open access: yesPhotodiagnosis and Photodynamic Therapy
While the colorimetric assay commonly referred to as ‘MTT’ does provide an indication of effects of photodamage on mitochondrial enzyme levels, a careful analysis showed that the resulting data were not correlated with a clonogenic assay.

Cannabidiol stimulates AML-1a-dependent glial differentiation and inhibits glioma stem-like cells proliferation by inducing autophagy in a TRPV2-dependent manner [PDF]

open access: yes, 2015
Glioma stem-like cells (GSCs) correspond to a tumor cell subpopulation, involved in glioblastoma multiforme (GBM) tumor ini- tiation and acquired chemoresistance.

Luteolin and Vernodalol as Bioactive Compounds of Leaf and Root Vernonia amygdalina Extracts: Effects on α-Glucosidase, Glycation, ROS, Cell Viability, and In Silico ADMET Parameters

open access: yesPharmaceutics, 2023
The aqueous decoctions of Vernonia amygdalina (VA) leaves and roots are widely used in traditional African medicine as an antidiabetic remedy. The amount of luteolin and vernodalol in leaf and root extracts was detected, and their role was studied ...

Elevated intracellular cAMP exacerbates vulnerability to oxidative stress in optic nerve head astrocytes. [PDF]

open access: yes, 2018
Glaucoma is characterized by a progressive loss of retinal ganglion cells and their axons, but the underlying biological basis for the accompanying neurodegeneration is not known.

The Effect of Bioceramic-Based Root Canal Sealers on the Cell Viability and Cytotoxicity of Human Periodontal Ligament Fibroblast cell line (An in vitro study) [PDF]

open access: yesAl-Rafidain Dental Journal
The current study aimed to assess the cell viability and cytotoxicity of bioceramic-based root canal sealers by comparing them to a zinc oxide-eugenol-based sealer (Endovit) in the set state of human periodontal ligament fibroblasts for one day, three ...

PARP inhibitors elicit distinct transcriptional programs in homologous recombination competent castration‐resistant prostate cancer

open access: yesMolecular Oncology, EarlyView.
PARP inhibitors are used to treat a small subset of prostate cancer patients. These studies reveal that PARP1 activity and expression are different between European American and African American prostate cancer tissue samples. Additionally, different PARP inhibitors cause unique and overlapping transcriptional changes, notably, p53 pathway upregulation.

MTT assay v1

open access: yes, 2022
The MTT assay is based on the conversion of MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide into purple formazan crystals by LIVINGcells, which determines the mitochondrial activity of these cells. Viable cells contain NADPH-dependent oxidoreductase enzymes which reduce MTT to formazan.1The insoluble formazan crystals ...

Adenosine‐to‐inosine editing of miR‐200b‐3p is associated with the progression of high‐grade serous ovarian cancer

open access: yesMolecular Oncology, EarlyView.
A‐to‐I editing of miRNAs, particularly miR‐200b‐3p, contributes to HGSOC progression by enhancing cancer cell proliferation, migration and 3D growth. The edited form is linked to poorer patient survival and the identification of novel molecular targets.
