Small-molecule targeting of MUSASHI RNA-binding activity in acute myeloid leukemia
The RNA binding protein MUSASHI-2 (MSI2) is a potential therapeutic target for acute myeloid leukemia. Here the authors identify a small molecule inhibitor of MSI2 and characterize its effects in a murine leukemia model.

A CLK3-HMGA2 Alternative Splicing Axis Impacts Human Hematopoietic Stem Cell Molecular Identity throughout Development [PDF]
While gene expression dynamics have been extensively cataloged during hematopoietic differentiation in the adult, less is known about transcriptome diversity of human hematopoietic stem cells (HSCs) during development. To characterize transcriptional and

RNA binding protein Musashi‐2 regulates PIWIL1 and TBX1 in mouse spermatogenesis
RNA‐binding proteins (RBP) are important facilitators of post‐transcriptional gene regulation. We have previously established that nuclear overexpression of the RBP Musashi‐2 (MSI2) during male germ cell maturation is detrimental to sperm cell development and fertility.

Hydrogen-fueled internal combustion engines [PDF]
The threat posed by climate change and the striving for security of energy supply are issues high on the political agenda these days. Governments are putting strate-gic plans in motion to decrease primary energy use, take carbon out of fuels and ...

HSCs: slow me down with PLAG1 [PDF]
In this issue of Blood, Keyvani Chahi et al1 have described the transcription factor PLAG1 as a positive regulator of human cord blood (CB) hematopoietic stem cell (HSC) dormancy and self-renewal by repressing the expression of translational machinery ...

Genome-wide analysis of Musashi-2 targets reveals novel functions in governing epithelial cell migration [PDF]
The Musashi-2 (Msi2) RNA-binding protein maintains stem cell self-renewal and promotes oncogenesis by enhancing cell proliferation in hematopoietic and gastrointestinal tissues. However, it is unclear how Msi2 recognizes and regulates mRNA targets in vivo and whether Msi2 primarily controls cell growth in all cell types. Here we identified Msi2 targets

We present a tissue‐stimulator platform for seamless electrode integration with pancreatic tissue, applying uniform electrical stimulation through optimized design with biohybrid 3D printing. Advantageous effects of electrical stimulation on β‐cell function were observed, including enhanced calcium signaling, islet morphology, and maturation.

An mRNA‐liposome vaccine via dissolving microneedles (ML‐DMN) enables efficient skin‐targeted delivery while preserving mRNA integrity and allowing sustained antigen expression. Unlike intramuscular (IM) injection, ML‐DMN engages skin‐resident immune cells, activates antigen‐presenting cells (APCs), and induces strong T cell and cytokine responses at ...
