Synthetic Ligands of Myeloid C‐Type Lectin Receptors
Binding of ligands to C‐type lectin receptors (CLRs) on myeloid cells can lead to an array of cellular responses. Exploitation of these receptors for therapeutic purposes requires high‐affinity ligands. This review surveys the diversity of synthetic ligands of myeloid CLRs in the context of binding, signaling, and downstream effects, as well as ...

Mycobacterium tuberculosis Rv3802c encodes a phospholipase/thioesterase and is inhibited by the antimycobacterial agent tetrahydrolipstatin. [PDF]
The cell wall of M. tuberculosis is central to its success as a pathogen. Mycolic acids are key components of this cell wall. The genes involved in joining the alpha and mero mycolates are located in a cluster, beginning with Rv3799c and extending at ...

Effects of ethambutol on accumulation and secretion of trehalose mycolates and free mycolic acid in Mycobacterium smegmatis [PDF]
We examined the early effects of ethambutol on the synthesis of trehalose monomycolate, trehalose dimycolate, and free mycolic acid in actively growing cells of Mycobacterium smegmatis. At about 1 min after the addition of 3.0 micrograms of ethambutol per ml, the cellular level of trehalose monomycolate began to increase over the control culture.

Machine learning–guided engineering of a plectasin‐derived peptide yields DC05, a potent antimycobacterial candidate. Encapsulation into tuftsin‐functionalized mesoporous silica nanoparticles enhances intracellular delivery, stability, and activity against Mycobacterium tuberculosis while maintaining low cytotoxicity and minimal hemolysis. The combined

This study employed multi‐omics approaches to investigate the regulatory mechanism of Mycobacterium PknG on host cell processes. We found that host lysine‐tRNA ligase (KARS) is a potential substrate of PknG; and PknG can regulate the immune response by catalyzing the phosphorylation of KARS at T592 and T595 sites, affecting the phosphorylation level in

Foamy macrophages from tuberculous patients' granulomas constitute a nutrient-rich reservoir for M. tuberculosis persistence. [PDF]
Tuberculosis (TB) is characterized by a tight interplay between Mycobacterium tuberculosis and host cells within granulomas. These cellular aggregates restrict bacterial spreading, but do not kill all the bacilli, which can persist for years.

ABSTRACT Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a leading global cause of death due to the pathogen's highly adaptive physiology and its ability to manipulate host immunity. Despite extensive study, our understanding of Mtb biology during infection remains incomplete, limiting the development of new diagnostics ...

Two Accessory Proteins Govern MmpL3 Mycolic Acid Transport in Mycobacteria
Mycolic acids are the signature lipid of mycobacteria and constitute an important physical component of the cell wall, a target of mycobacterium-specific antibiotics and a mediator of Mycobacterium tuberculosis pathogenesis. Mycolic acids are synthesized

Uncovering Key Characteristics of Antibacterial Peptides through Machine Learning
Machine‐learning (ML) techniques using random forest classification models revealed key characteristics that predict effective antimicrobial peptides (AMPs) targeting Gram‐negative bacteria, Gram‐positive bacteria, and mycobacteria. The ideal cLogP (<$ < $−6) and net‐charge (≤+4) threshold was the same for all three targets with variations in the ...
