Use of cultured airway myocytes for study of airway smooth muscle

American Journal of Physiology-Lung Cellular and Molecular Physiology, 1995
Cultured airway smooth muscle cells provide a convenient model system for studying the regulation of a wide range of airway responses at the cellular level. This review describes the characteristics of cultured airway smooth muscle cells and differences that exist between cultured cells and acutely dissociated cells or muscle strips.

Responses of subcultured rat aortic smooth muscle myocytes to vasoactive agents and KCl-induced depolarization

American Journal of Physiology-Cell Physiology, 1991
We have developed a culture system in which a single-mass primary culture can be used for as long as 6 wk as a source of subcultured smooth muscle myocytes for the study of the changes of their shape upon addition of vasoactive agents (angiotensin, vasopressin, norepinephrine, and serotonin) and KCl depolarization.

Identification of a novel isoform of the cyclic-nucleotide phosphodiesterase PDE3A expressed in vascular smooth-muscle myocytes

open access: yesBiochemical Journal, 2000
We have identified a new cyclic-nucleotide phosphodiesterase isoform, PDE3A, and cloned its cDNA from cultured aortic myocytes. The nucleotide sequence of its coding region is similar to that of the previously cloned myocardial isoform except for the absence of the initial 300–400nt that are present in the latter, as confirmed by reverse-transcriptase ...

High-efficiency somatic mutagenesis in smooth muscle cells and cardiac myocytes in SM22?-Cre transgenic mice

genesis, 2005
The cytoskeletal protein SM22alpha is expressed in visceral and vascular smooth muscle cells (SMCs), in cardiac myocytes, and in the myotomal components of the somites during murine embryonic development. In this report, we describe the generation and characterization of transgenic mice expressing Cre-recombinase under the transcriptional control of ...

Altered Ion Channel Activity in Murine Colonic Smooth Muscle Myocytes in an Experimental Colitis Model

Biochemical and Biophysical Research Communications, 2000
We have investigated the activity of calcium and potassium channels in a murine model of experimental colitis. Colonic myocytes from dextran sulphate sodium (DSS)-treated mice were examined by whole cell patch clamp techniques. Myeloperoxidase activity was enhanced 3. 5-fold in DSS-treated mouse colon.

The role of mechanical and humoral factors in growth regulation of vascular smooth muscle and cardiac myocytes

Current Opinion in Nephrology and Hypertension, 1993
Structural changes of the heart and blood vessels participate in the long-term regulation of the cardiovascular system. In hypertension and myocardial dysfunction, the adaptive process of cardiac and vascular remodeling may contribute to the pathophysiology and complications of these diseases.

Overexpression of human Hsp27 inhibits serum-induced proliferation in airway smooth muscle myocytes and confers resistance to hydrogen peroxide cytotoxicity [PDF]

open access: possibleAmerican Journal of Physiology-Lung Cellular and Molecular Physiology, 2007
Airway smooth muscle (ASM) hypertrophy and hyperplasia are characteristics of asthma that lead to thickening of the airway wall and obstruction of airflow. Very little is known about mechanisms underlying ASM remodeling, but in vascular smooth muscle, it is known that progression of atherosclerosis depends on the balance of myocyte proliferation and ...

Synthesis and Expression of Tissue Factor Pathway Inhibitor by Serum-stimulated Fibroblasts, Vascular Smooth Muscle Cells and Cardiac Myocytes

Thrombosis and Haemostasis, 1999
SummaryTissue factor pathway inhibitor (TFPI) plays an important role in regulating tissue factor (TF)-initiated blood coagulation. Since serum stimulation of fibroblasts, vascular smooth muscle cells and cardiac myocytes in culture increases the expression of TF mRNA and antigen (Ag) in these cells, we hypothesized that serum may also induce increased

Hypertension Differentially Affects the Expression of the Gap Junction Protein Connexin43 in Cardiac Myocytes and Aortic Smooth Muscle Cells

Advances in Experimental Medicine and Biology, 1997
Electrical and mechanical coupling of myocytes in heart and of smooth muscle cells in the aortic wall is thought to be mediated by intercellular channels aggregated at gap junctions. Connexin43 (Cx43) is one of the predominant membrane proteins forming junctional channels in the cardiovascular system.

Adenovirus-mediated Overexpression of Human Transforming Growth Factor-β1 in Rat Cardiac Fibroblasts, Myocytes and Smooth Muscle Cells

Journal of Molecular and Cellular Cardiology, 1996
Transforming growth factor-beta 1 (TGF-beta 1) is known to regulate cardiac cell function and its overexpression in the heart is thought to contribute to the development of cardiac hypertrophy and fibrosis. We wished to develop a high efficiency gene transfer method that could be used both in vitro and in vivo and result in the overexpression of TGF ...
