Mycobacterial cell division arrest and smooth‐to‐rough envelope transition using CRISPRi‐mediated genetic repression systems

open access: yesFEBS Open Bio, EarlyView.
CRISPRI‐mediated gene silencing and phenotypic exploration in nontuberculous mycobacteria. In this Research Protocol, we describe approaches to control, monitor, and quantitatively assess CRISPRI‐mediated gene silencing in M. smegmatis and M. abscessus model organisms.

Exon 7 splicing of ERα predicts poor prognosis and increases phenotypic heterogeneity in luminal a subtype breast cancer

open access: yesFEBS Open Bio, EarlyView.
ERα splice variant ERα∆7 lacks the C‐terminus, and its expression may change phenotypes of breast cancers. Our results showed that ERα∆7 is found in the luminal A subtype, and elevated ERα∆7 levels are linked to improved cell survival with lower proliferation and migration.

Morphological Classification of 52 Accessions of White Yam (Dioscorea rotundata) Species at the National Root Crops Research Institute, Umudike, Abia State, Nigeria

open access: yesJournal of Applied Sciences and Environmental Management
The classification and identification of white yam (Dioscorea rotundata) species are often confusing and poorly characterized, hindering breeding programmes and conservation efforts. The objective of this paper was classification of 52 Accessions of white yam (Dioscorea rotundata) species at African yam screen house, National Root Crops Research ...

Screening and epitope characterization of Nidogen‐2‐specific nanobodies

open access: yesFEBS Open Bio, EarlyView.
Camel immunization and phage display were employed to generate high‐affinity VHH nanobodies against Nidogen‐2. After library construction, biopanning, ELISA screening, sequencing, and recombinant expression, selected nanobodies were purified and characterized, leading to the preliminary exploration of a nanobody‐based sandwich ELISA for specific ...

Genotype-by-environment interaction and stability of root mealiness and other organoleptic properties of boiled cassava roots

open access: yesScientific Reports, 2022
Genetic enhancement of cassava aimed at improving cooking and eating quality traits is a major goal for cassava breeders to address the demand for varieties that are desirable for the fresh consumption market segment.

Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.

Novel SNP markers and other stress-related genomic regions associated with nitrogen use efficiency in cassava

open access: yesFrontiers in Plant Science
Cassava productivity is constrained by low soil nitrogen, which is predominant in most cassava-growing regions in the tropics and subtropical agroecology. Improving the low nitrogen tolerance of cassava has become an important breeding objective.

DESIGN OF SOLAR PHOTOVOLTAIC SYSTEM FOR UNDERGROUND IRRIGATION PUMPING WATER FOR THE AGRICULTURAL FARMLAND IN NATIONAL ROOT CROPS RESEARCH INSTITUTE, UMUDIKE (NRCRI), ABIA STATE, NIGERIA.

open access: yesJournal of Science and Technology
This study addresses the critical challenge of water scarcity and high energy costs impacting agricultural productivity in regions like Abia State, Nigeria, by proposing a sustainable solar photovoltaic (SPV) irrigation pumping system. The objective was to design and evaluate an SPV system for underground irrigation at the National Root Crops Research ...

Mass spectrometry based identification of AMP‐O‐Tris generated by Thermococcus onnurineus Cas10

open access: yesFEBS Open Bio, EarlyView.
Isolated Thermococcus onnurineus Cas10 generates the noncanonical ATP‐derived product AMP‐O‐Tris while in Tris‐containing buffer as identified via mass spectrometry, revealing relaxed nucleophile selectivity under isolated conditions. These findings suggest that multiprotein Csm complex assembly restricts Cas10 reactivity toward canonical cyclic ...
