Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Metastasis on pause: How dormant tumor cells stay hidden within the tumor microenvironment and evade immune surveillance

open access: yesMolecular Oncology, EarlyView.
Dormant cancer cells can hide in distant organs for years, evading treatment and the immune system. This review highlights how signals from the surrounding tissue and immune environment keep these cells inactive or trigger their reawakening. Understanding these mechanisms may help develop therapies to eliminate or control dormant cells and prevent ...

Time in and for nature-based solutions. No quick fix solutions for complex ecological and social processes

open access: yesNature-Based Solutions
Nature-based solutions (NbS) are an increasingly widespread tool in environment and development policies. They embody the idea that nature and natural processes can be purposefully used by humans to provide solutions to pressing environmental and social ...

DNA methylation and expression of MAPRE3 affect overall survival of early‐stage non‐small cell lung cancer patients

open access: yesMolecular Oncology, EarlyView.
Both cg12821679MAPRE3 methylation and MAPRE3 expression are significantly associated with overall survival (OS) of non‐small cell lung cancer. Meanwhile, MAPRE3 expression significantly modified the effect of smoking cessation on OS. Smoking cessation benefits OS merely for patients with high MAPRE3 expression.

Effect of compost and compost-derived biochar on the growth of lettuce irrigated with water and treated wastewater

open access: yesNature-Based Solutions
As climate change keeps progressing, the use of biochar and compost as nature-based materials emerges as sustainable alternative that can have a positive impact on agriculture.

Loss of IGF‐1R impairs DNA‐PKcs recruitment to chromatin leading to defective end‐joining

open access: yesMolecular Oncology, EarlyView.
IGF‐1R promotes radioresistance by facilitating DNA‐PKcs recruitment to chromatin, enabling non‐homologous end‐joining (NHEJ) repair of double‐strand breaks. Inhibition or loss of IGF‐1R disrupts this recruitment to damage sites, driving compensatory reliance on microhomology‐mediated end‐joining (MMEJ) repair.

Advancing blue-green infrastructure design with synthetic 3D drainage channels: A scenario-based flood model in Nova Scotia, Canada

open access: yesNature-Based Solutions
Urbanized riverscapes are facing challenges due to hydrological changes. Adjusted flow regimes and imperviousness are contributing to increased flood risks resulting from gray infrastructure and strained subgrade drainage systems.

Adoption of nature-based solutions and orchard sustainable management to face kiwifruit vine decline syndrome (KVDS)

open access: yes, 2022
Italy, the third largest producers of kiwifruit in the world, lost 10% of its production in recent years because of the spread of the kiwifruit vine decline syndrome (KVDS) (Bardi, 2020).
