The ionization constant of nicotinamide-adenine dinucleotide

Analytical Biochemistry, 1969
The pKa of carefully purified oxidized nicotinamide adenine dinucleotide has been determined by potentiometric titration with sodium hydroxide at 25°. Values are reported for six ionic strengths up to 0.250. Extrapolation to zero ionic strength yields 3.88 ± 0.02 for the thermodynamic pKa.

Thermodynamics of Reactions of Nicotinamide Adenine Dinucleotide and Nicotinamide Adenine Dinucleotide Phosphate

Archives of Biochemistry and Biophysics, 1993
The thermodynamics of six reactions of nicotinamide adenine dinucleotide and nicotinamide adenine dinucleotide phosphate is discussed both from the viewpoint of the chemical equations and the biochemical equations for these reactions. Tables of the standard enthalpies of formation and standard Gibbs energies of formation of species are presented and ...

Psychopharmacological action of nicotinamide adenine dinucleotide

Psychopharmacologia, 1970
The effect of nicotinamide adenine dinucleotide (NAD) upon the response rates in variable interval and Sidman Avoidance schedules was studied in rats. Bar pressing on the variable interval reinforcement schedule was significantly reduced during the 15 min following the administration of 10 mg/kg i.p.

Electrochemical regeneration of nicotinamide adenine dinucleotide

Biochimica et Biophysica Acta (BBA) - General Subjects, 1975
Reduced nicotinamide adenine dinucleotide (NADH) has been characterized electrochemically by solid electrode voltammetry and controlled potential electrolysis. Photometric and enzymatic assay showed that enzymatically active nicotinamide adenine dinucleotide (NAD-+) could be regenerated electrolytically from its reduced form without the use of so ...

The biosynthesis of nicotinamide adenine dinucleotides in bacteria

2001
The nicotinamide adenine dinucleotides (NAD, NADH, NADP, and NADPH) are essential cofactors in all living systems and function as hydride acceptors (NAD, NADP) and hydride donors (NADH, NADPH) in biochemical redox reactions. The six-step bacterial biosynthetic pathway begins with the oxidation of aspartate to iminosuccinic acid, which is then condensed

Liquid chromatographic separations of dimers of nicotinamide adenine dinucleotide and nicotinamide adenine dinucleotide phosphate

Journal of Chromatography A, 1985
Abstract The dimers of nicotinamide adenine dinucleotide (NAD) and nicotinamide adenine dinucleotide phosphate (NADP) [(NAD)2 and (NADP)2, respectively[ were prepared electrolytically from NAD and NADP and analysed by several chromatographic techniques. Gel permeation chromatography on UltroPac columns was more convenient for the separation of dimers

Selective inhibition of nicotinamide adenine dinucleotide kinases by dinucleoside disulfide mimics of nicotinamide adenine dinucleotide analogues

Bioorganic & Medicinal Chemistry, 2009
Diadenosine disulfide (5) was reported to inhibit NAD kinase from Listeria monocytogenes and the crystal structure of the enzyme-inhibitor complex has been solved. We have synthesized tiazofurin adenosine disulfide (4) and the disulfide 5, and found that these compounds were moderate inhibitors of human NAD kinase (IC(50)=110 microM and IC(50)=87 ...

Nicotinamide Adenine Dinucleotide Based Therapeutics, Update

Current Medicinal Chemistry, 2015
About 500 NAD (P)-dependent enzymes in the cell use NAD (P) as a cofactor or a substrate. This family of broadly diversified enzymes is crucial for maintaining homeostasis of all living organisms. The NAD binding domain of these enzymes is conserved and it was believed that NAD mimics would not be of therapeutic value due to lack of selectivity ...

Nicotinamide-adenine dinucleotide pyrophosphatase of Cambaroides japonica

Biochimica et Biophysica Acta (BBA) - Enzymology, 1969
Abstract 1. 1.The cleavage of NAD in crayfish hepatopancreas is catalyzed chiefly by a pyrophosphatase rather than by NAD glycohydrolase (EC 3.2.2.5). This fact was confirmed by the loss in the coenzyme function of NAD for yeast alcohol dehydrogenase without a significant concomitant loss in reactivity towards cyanide and the identification of ...
