Nicotinamide Phosphoribosyltransferase/Visfatin Does Not Catalyze Nicotinamide Mononucleotide Formation in Blood Plasma

open access: yesNicotinamide Phosphoribosyltransferase/Visfatin Does Not Catalyze Nicotinamide Mononucleotide Formation in Blood Plasma
Nicotinamide (Nam) phosphoribosyltransferase (NAMPT) is the rate-limiting enzyme in mammalian NAD synthesis, catalyzing nicotinamide mononucleotide (NMN) formation from Nam and 5-phosphoribosyl 1-pyrophosphate (PRPP). NAMPT has also been described as an adipocytokine visfatin with a variety of actions, although physiological significance of this ...

Medicinal Chemistry of Nicotinamide Phosphoribosyltransferase (NAMPT) Inhibitors

Journal of Medicinal Chemistry, 2013
Nicotinamide phoshophoribosyltransferase (NAMPT) plays a key role in the replenishment of the NAD pool in cells. This in turn makes this enzyme an important player in bioenergetics and in the regulation of NAD-using enzymes, such as PARPs and sirtuins. Furthermore, there is now ample evidence that NAMPT is secreted and has a role as a cytokine.

A rapid procedure for assaying nicotinamide phosphoribosyltransferase

Analytical Biochemistry, 1980
Abstract A precipitation-filtration assay which measures the activity of nicotinamide phosphoribosyltransferase is described. The assay, based on the differential solubility of nicotinamide and nicotinamide mononucleotide in acetone, is rapid, convenient, and inexpensive when compared to chromatographic methods.

Nicotinamide phosphoribosyltransferase regulates cocaine reward through Sirtuin 1

Experimental Neurology, 2018
Nicotinamide phosphoribosyltransferase (NAMPT), a rate-limiting enzyme in nicotinamide adenine dinucleotide (NAD) biosynthesis in mammals, converts nicotinamide into nicotinamide mononucleotide (NMN). NMN is subsequently converted to NAD, a component that is critical for cell energy metabolism and survival.

Nicotinamide phosphoribosyltransferase (Nampt) in carcinogenesis: new clinical opportunities

Expert Review of Anticancer Therapy, 2016
Nicotinamide phosphoribosyltransferase (Nampt) is the rate-limiting enzyme that catalyzes the first step in the mammalian nicotinamide adenine dinucleotide (NAD) salvage pathway. Aberrant NAD metabolism was associated with oncogenic signal transduction, suggesting the critical roles of Nampt in tumorigenesis and metastasis.
