Focusing on the discovery of new therapeutics for childhood cancer, novel quinoline derivatives were synthesized and demonstrated activity against Ewing sarcoma. A target search was performed against a panel of kinases, and the compounds showed selectivity for the NAK family, mainly GAK, which was confirmed by enzyme kinetic assays.

Three novel Ln(III) complexes for efficient, site‐specific cysteine labeling under mild aqueous conditions are reported. A kinetic study reveals how subtle changes to the reactive pendant arm tune tagging rates, with Tb.QuNO achieving >95% cysteine labeling in 10 min.

Investigation of the structure‐activity relationship on W482 (1) with a pyrimidine‐2,4‐diamine scaffold culminated in analog 51 with enhanced anti‐plasmodial activity. Forward genetics and phenotypic examination revealed the ABCI3 transporter protein as a putative resistance mechanism.

Our work demonstrates that time‐dependent auxiliary density functional theory (TD‐ADFT) in combination with the hybrid diagonal approximation (HDA) for global and range‐separated hybrid functionals is a promising approach for low‐order scaling electronic excitation energy calculations.
