Vasculitis and the NLRP3 inflammasome
Current Opinion in Rheumatology, 2023Purpose of review Vasculitis are a group of heterogeneous conditions characterized by chronic inflammation of blood vessels, leading to tissue destruction and organ failure. Vasculitis is an inflammatory process in which immune effector cells infiltrate blood vessels and surrounding tissues.

NLRP3 inflammasome in neurodegenerative disease
Translational Research, 2023Neurodegenerative diseases are characterized by a dysregulated neuro-glial microenvironment, culminating in functional deficits resulting from neuronal cell death. Inflammation is a hallmark of the neurodegenerative microenvironment and despite a critical role in tissue homeostasis, increasing evidence suggests that chronic inflammatory insult can ...

NLRP3 inflammasome inhibitor INF39 attenuated NLRP3 assembly in macrophages
International Immunopharmacology, 2021INF39 is a nontoxic, irreversible, acrylate-based NLRP3 inhibitor and a further optimization of ethyl 2-((2-chlorophenyl) hydroxyl) methyl) acrylate (INF4E). However, the detail mechanism and the direct target of its anti-inflammatory activity is not clear. Here, we show that INF39 is a specific inhibitor for NLRP3 inflammasome activation.

NLRP3 inflammasome activation after ischemic stroke
Behavioural Brain Research, 2023Cerebral ischemia is a pathological condition resulting from the cessation or reduction of blood supply to the cerebral arteries. Neurological deficits that are clinically relevant can arise as a result of brain damage. The etiology of stroke is multifaceted and intricate, with the inflammatory response being a crucial component that warrants ...

Leukadherin-1 inhibits NLRP3 inflammasome by blocking inflammasome assembly
International Immunopharmacology, 2023Aberrant activation of the NLRP3 inflammasome has been implicated in the occurrence and development of many inflammatory diseases, and thus potent inhibitors of the NLRP3 inflammasome should be explored. An antitumor agent, Leukadherin-1 (LA-1), tested in phase 1/2 clinical trials, has been reported to exert anti-inflammatory properties by blocking the

The Nlrp3 inflammasome admits defeat
Trends in Immunology, 2015The Nlrp3 inflammasome triggers interleukin-1 secretion by myeloid cells in response to endogenous and exogenous danger signals. Two recent studies identified the sulfonylurea MCC950 and the ketone metabolite β-hydroxybutyrate as specific inhibitors of the Nlrp3 inflammasome, with promising therapeutic potential for the treatment of auto-inflammatory ...

Methylsulfonylmethane inhibits NLRP3 inflammasome activation
Cytokine, 2015Methylsulfonylmethane (MSM) is an organosulfur compound and the health benefits associated with MSM include inflammation. Although MSM has been shown to have various physiological effects, no study has yet focused on inflammasome activation. The inflammasome is a multiprotein complex that serves as a platform for caspase 1-dependent proteolytic ...

The NLRP3 inflammasome: activation and regulation
Trends in Biochemical Sciences, 2023The NOD-, LRR- and pyrin domain-containing protein 3 (NLRP3) inflammasome is a cytoplasmic supramolecular complex that is activated in response to cellular perturbations triggered by infection and sterile injury. Assembly of the NLRP3 inflammasome leads to activation of caspase-1, which induces the maturation and release of interleukin-1β (IL-1β) and ...

NLRP3 inflammasome activation in inflammaging
Seminars in Immunology, 2018The process of aging is associated with the appearance of low-grade subclinical inflammation, termed inflammaging, that can accelerate age-related diseases. In Western societies the age-related inflammatory response can additionally be aggravated by an inflammatory response related to modern lifestyles and excess calorie consumption, a pathophysiologic

NLRP3 Inflammasome in Acute Myocardial Infarction
Journal of Cardiovascular Pharmacology, 2019Abstract:Acute myocardial infarction (AMI) is associated with the induction of a sterile inflammatory response that leads to further injury. The NACHT, leucine-rich repeat, and pyrin domain–containing protein 3 (NLRP3) inflammasome is a macromolecular structure responsible for the inflammatory response to injury or infection.
