Celastrol upregulates adipocyte‐derived exosomal miR‐5099, suppressing the macrophage c‐Met/NF‐κB axis to mitigate pro‐inflammatory M1 polarization and adipose inflammation. These miR‐5099‐enriched exosomes also act as endocrine signals targeting the liver, muscle, and adipocytes to significantly enhance systemic insulin sensitivity.

NIBAN2/FLII/RREB1 Axis Drives Glioma Stem Cell Malignancy via TLR3 Pathway Activation
NIBAN2, highly expressed in glioma stem‐like cells (GSCs), assembles with FLII and transcription factor RREB1 to form a nuclear complex. This complex transcriptionally activates stemness‐associated genes (e.g., CD44, NANOG) and metabolic enzymes (e.g., LDHA), thereby sustaining both transcriptional and metabolic stemness programs.

Cellular Identity Crisis: RD3 Loss Fuels Plasticity and Immune Silence in Progressive Neuroblastoma
Researchers discovered that therapy‐induced loss of RD3 protein in neuroblastoma triggers a dangerous shift: cancer cells become more stem‐like, invasive, and resistant to treatment while evading immune detection. The results showed that the reduction of plasma 24‐hydroxycholesterol (24‐OHC) was related to the cognitive decline in elderly surgical patients. Neuroinflammation induced by anesthesia and surgery significantly downregulated SREBP2 in hippocampal reactive astrocytes, resulting in reduced cholesterol synthesis.

We studied how five common immunosuppressants behave when delivered directly to a transplant site instead of systemically. Using a vascularized implant for islet transplantation, we show that local delivery protects grafts, limits drug spread to the rest of the body, and produces distinct immune signatures.
