Harnessing changes in cellular glycosylation in new cancer treatment strategies.

open access: yes, 2004
The majority of proteins are modified in post-translational events and one of the most common of these is glycosylation. Many reports describe alterations to the normal cellular glycosylation in cancer but detailed knowledge of the underlying structures ...

Incorporation of Novel Synthetic Glycolipids in Liposomal Nanoparticles Affects Opsonization and In Vivo Clearance

open access: yesAngewandte Chemie, EarlyView.
We prepared five glycosylated liposomal nanoparticles (G‐LNPs) to investigate the role of glycosylation and protein corona in modulating the in vivo behavior of G‐LNPs. We show that IgG and complement C3 adsorption enhanced liposomal nanoparticle clearance, with IgG promoting subsequent C3 binding.

Total Chemical Synthesis of Interleukin‐15 and Interleukin‐2: Taming Protein Hydrophobicity and Aggregation by a Versatile Solubilizing Strategy

open access: yesAngewandte Chemie, EarlyView.
A reducible solubilizing tag strategy (RST‐2.0) enables the synthesis, purification and ligation of difficult peptides while being readily removable just during folding, which overcomes the common challenges from protein hydrophobicity and aggregation. With this method, we achieved the first total synthesis of native and N‐glycosylated IL‐15 as well as

Fluorogenic Coupled Assays Reveal Catalytic Properties, Inhibition Constants and Cellular Location of Mucin‐Active Carbohydrate Sulfatases

open access: yesAngewandte Chemie, EarlyView.
We report plate‐based coupled assays to rapidly and quantitatively determine the activity and selectivity of mucin‐active carbohydrate sulfatases. This fluorogenic assay further enables inhibitor identification and allows determination of (sub‐)cellular location. ABSTRACT Sulfated glycans play a central role in human health and influence cell signaling,

Expanding the Utility of Exome Sequencing in Preventive and Population Genetics

open access: yesAmerican Journal of Medical Genetics Part A, EarlyView.
ABSTRACT Carrier screening is a long‐standing genetic testing process offered to at‐risk couples, with or without a family history, who might have pregnancies affected by an autosomal recessive (AR) or X‐linked (XL) disorder. A total of 276 unrelated individuals, initially referred for rare disorder screening by clinicians, were enrolled in this study ...

3’-O-β-Glycosylation of nucleoside analogues using a promiscuous bacterial glycosyltransferase

open access: yes
Nucleoside analogue therapeutics have a proven capability within drug discovery as antimicrobial, antiviral and antineoplastic agents. However, their efficacy can be limited by poor cellular uptake, high off target toxicity and poor bioavailability ...

Carbohydrate Physicochemical Properties: The Innate Hydrogen Bond Donating Capacities of α‐Glucoside and α‐Galactoside Alcohol Groups

open access: yesAngewandte Chemie, EarlyView.
Experimental determination of the innate hydrogen bond donating capacities of the glucose and galactose non‐anomeric alcohols revealed significant variations depending on position and relative configuration, which were quantified on the pKAHY scale, relevant for MedChem purposes, hence providing another tool toward rational glycomimetic development ...
