Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

A study on the convergence of observer-based kinetics estimators in stirred tank bioreactors [PDF]

open access: yes, 1994
This paper is devoted to the tuning problem of the "observer-based kinetics estimator" in stirred tank bioreactors. This algorithm estimates the reaction kinetics from the on-line knowledge of the state variables (either from measurement or by means of

Gut microbiome and aging—A dynamic interplay of microbes, metabolites, and the immune system

open access: yesFEBS Letters, EarlyView.
Age‐dependent shifts in microbial communities engender shifts in microbial metabolite profiles. These in turn drive shifts in barrier surface permeability of the gut and brain and induce immune activation. When paired with preexisting age‐related chronic inflammation this increases the risk of neuroinflammation and neurodegenerative diseases.

A New Observer Design for the Joint Estimation of States and Unknown Inputs for a Class of Nonlinear Fractional-Order Systems

open access: yesMathematics
This article designs an observer for the joint estimation of the state and the unknown input for a class of nonlinear fractional-order systems (FOSs) such that one portion satisfies the Lipschitz condition and the other does not necessarily satisfy such ...

New Nonlinear Takagi–Sugeno Vehicle Model for State and Road Curvature Estimation via a Nonlinear PMI Observer

open access: yesJournal of Intelligent Systems, 2014
The present article deals with an observer design for nonlinear vehicle lateral dynamics. The contributions of the article concern the nonconsideration of any force model and the consideration that the longitudinal velocity is time varying, which is more

A methionine‐lined active site governs carbocation stabilization and product specificity in a bacterial terpene synthase

open access: yesFEBS Letters, EarlyView.
This study reveals a unique active site enriched in methionine residues and demonstrates that these residues play a critical role by stabilizing carbocation intermediates through novel sulfur–cation interactions. Structure‐guided mutagenesis further revealed variants with significantly altered product profiles, enhancing pseudopterosin formation. These

Observers for compressible Navier-Stokes equation

open access: yes, 2016
We consider a multi-dimensional model of a compressible fluid in a bounded domain. We want to estimate the density and velocity of the fluid, based on the observations for only velocity.

Plasma extrachromosomal circular DNA as a biomarker in EGFR‐targeted therapy of non‐small cell lung cancer

open access: yesMolecular Oncology, EarlyView.
Detection of extrachromosomal circular DNA (eccDNA) in plasma samples from EGFR‐mutated non‐small cell lung cancer patients. Plasma was collected before and during treatment with the EGFR‐tyrosine kinase inhibitor osimertinib. Plasma eccDNA was detected in all cancer samples, and the presence of the EGFR gene on eccDNA serves as a potential biomarker ...

Estimation of Sporulated Cell Concentration of Bacillus thuringiensis in a Batch Biochemical Reactor via Simple State Observers

open access: yesMathematics
This paper presents a contrast of two different observation strategies viz a nonlinear observer and a classical extended Luenberger observer applied to a bioreactor system for Bacillus thuringiensis production.

Dual targeting of RET and SRC synergizes in RET fusion‐positive cancer cells

open access: yesMolecular Oncology, EarlyView.
Despite the strong activity of selective RET tyrosine kinase inhibitors (TKIs), resistance of RET fusion‐positive (RET+) lung cancer and thyroid cancer frequently occurs and is mainly driven by RET‐independent bypass mechanisms. Son et al. show that SRC TKIs significantly inhibit PAK and AKT survival signaling and enhance the efficacy of RET TKIs in ...
