Epigenetic blind spots – the role of DNA methylation dynamics in stem cell‐based models of embryogenesis

open access: yesFEBS Letters, EarlyView.
Embryo‐like structures (stembryos) are an innovative tool, but they are hindered by experimental variability and limited developmental potential. DNA methylation is crucial for mammalian development, but its status in stembryo models is poorly characterized.

Reconstructing enzyme evolution by protein engineering

open access: yesFEBS Letters, EarlyView.
Natural enzyme evolution can be retraced by protein engineering methods such as directed evolution, rational design, and ancestral sequence reconstruction. These approaches reveal how enzymes emerged from ligand‐binding scaffolds, developed varying substrate preferences, formed oligomeric complexes, adapted to environmental changes, and evolved novel ...

Microplastics in Malaysia's Aquatic Environment: Current Overview and Future Perspectives

open access: yesGlobal Challenges, 2023
Microplastic pollution has adversely affected the aquatic ecosystem, living creatures, and human health. Several studies in Malaysia have provided baseline information on the existence of microplastics in surface water, ingestion by marine life and ...

The role of miR‐335‐5p in the redifferentiation of BRAF p.V600E thyroid cancers

open access: yesMolecular Oncology, EarlyView.
The BRAF p.V600E mutation promotes thyroid cancer dedifferentiation and radioiodine resistance. Using a network approach, we identified miR‐335‐5p as a key regulator of BRAF‐mutated thyroid tumors. Restoring miR‐335‐5p increased thyroid‐specific gene expression and iodine uptake in cells and organoids.

Somatic mutational landscape in von Hippel–Lindau familial hemangioblastoma

open access: yesMolecular Oncology, EarlyView.
The causes of central nervous system (CNS) hemangioblastoma in Von Hippel–Lindau (vHL) disease are unclear. We used Whole Exome Sequencing (WES) on familial hemangioblastoma to investigate events that underlie tumor development. Our findings suggest that VHL loss creates a permissive environment for tumor formation, while additional alterations ...

Dissecting and validation the biomarker of heart failure progression in patients with atherosclerosis by single-cell sequencing, bioinformatics, and machine learning

open access: yesFrontiers in Genetics
ObjectiveThis study aimed to identify early biomarkers associated with the progression from atherosclerosis (AS) to heart failure (HF) by integrating single-cell RNA sequencing (scRNA-seq) and bulk transcriptomic data, and to explore the potential ...

Differential expression of cancer‐related genes supports prediction of poor response to first‐line treatments in T‐ALL pediatric patients with high minimal residual disease

open access: yesMolecular Oncology, EarlyView.
In the present work, we have identified a transcriptional signature based on the differential expression of six genes (BCL2&MAST4, HSH2D&LAT2, METRN&PITPNM2) that would facilitate the early detection of T‐cell acute lymphoblastic leukemia (T‐ALL) patients prone to a poor treatment response and could be implemented at diagnosis, along with other risk ...

Performance changes during and post-exercise in combined heat and hypoxia environments: a systematic review and network meta analysis

open access: yesJournal of Translational Medicine
Background This study aims to compare exercise performance, as well as physiological, and psychological changes, when individuals exercise both during and after exposure to the heat environment (HEAT), the hypoxia environment (HYP), and the combined heat

Case Report: Uterine mesothelial cyst: a report of three cases and literature review

open access: yesFrontiers in Medicine
ObjectiveMesothelioma cysts are benign tumor-like lesions derived from mesothelial cells. They mainly affect the peritoneum of the pelvic and abdominal cavities, and are more commonly found in the mesentery and greater omentum, while they are less likely

CCDC80 suppresses high‐grade serous ovarian cancer migration via negative regulation of B7‐H3

open access: yesMolecular Oncology, EarlyView.
PAX8 is a lineage‐specific master regulator of transcription in high‐grade serous ovarian cancer (HGSC) progression. We show for the first time that PAX8 facilitates proliferation and metastasis by repressing the cell autonomous tumor suppressor CCDC80 and inducing B7‐H3 expression.
