Regulation of autophagy by SARS‐CoV‐2: The multifunctional contributions of ORF3a

Journal of Medical Virology, 2023
AbstractSevere acute respiratory syndrome‐coronavirus‐2 (SARS‐CoV‐2) regulates autophagic flux by blocking the fusion of autophagosomes with lysosomes, causing the accumulation of membranous vesicles for replication. Multiple SARS‐CoV‐2 proteins regulate autophagy with significant roles attributed to ORF3a. Mechanistically, open reading frame 3a (ORF3a)

Molecular conservation and differential mutation on ORF3a gene in Indian SARS-CoV2 genomes

open access: yesGenomics, 2020
Abstract A global emergency due to the COVID-19 pandemic demands various studies related to genes and genomes of the SARS-CoV2. Among other important proteins, the role of accessory proteins are of immense importance in replication, regulation of infections of the coronavirus in the hosts. The largest accessory proteins in the SARS-CoV2

SARS-CoV-2 ORF3a induces RETREG1/FAM134B-dependent reticulophagy and triggers sequential ER stress and inflammatory responses during SARS-CoV-2 infection

open access: yesAutophagy, 2022
SARS-CoV-2 infections have resulted in a very large number of severe cases of COVID-19 and deaths worldwide. However, knowledge of SARS-CoV-2 infection, pathogenesis and therapy remains limited, emphasizing the urgent need for fundamental studies and ...

Investigating SARS-CoV-2 ORF3a binding to human TRAF2 and TRAF3

The Journal of Immunology, 2023
Abstract Severe COVID-19, caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), is characterized by widespread inflammation and cytokine storm. A number of viral proteins are capable of inducing inflammatory responses, one of which is open reading frame 3a (ORF3a). ORF3a from SARS-CoV shares 73% homology with SARS-CoV-2

Μελέτη της πρωτεΐνης ORF3a του SARS-CoV-2

2022

Genetic variety of ORF3a shapes SARS‐CoV‐2 fitness through modulation of lipid droplet

Journal of Medical Virology, 2023
AbstractSevere acute respiratory syndrome coronavirus‐2 (SARS‐CoV‐2) infection leads to the accumulation of lipid droplets (LD), the central hubs of the lipid metabolism, in vitro or in type II pneumocytes and monocytes from coronavirus disease 19 (COVID‐19) patients and blockage of LD formation by specific inhibitors impedes SARS‐CoV‐2 replication ...

Analysis of SARS-CoV-2 ORF3a structure reveals chloride binding sites

2020
Abstract SARS-CoV-2 ORF3a is believed to form ion channels, which may be involved in the modulation of virus release, and has been implicated in various cellular processes like the up-regulation of fibrinogen expression in lung epithelial cells, downregulation of type 1 interferon receptor, caspase-dependent apoptosis, and increasing ...

Impact of ORF3a-Q57H variant of SARS-CoV-2 on apoptotic signaling cascades

Physiology, 2023
Introduction: Direct infection and viral protein expression of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in the respiratory system, and other organs including the heart, contribute to multi-organ dysfunction during COVID-19. We previously showed that the protein encoded by the open reading frame 3a (ORF3a) gene of SARS-CoV-2, which ...

Insights into Ion Conduction Mechanisms Through the ORF3a Channel by Computational Modelling

Investigating the selectivity and mechanism of the SARSCoV-2 ORF3a protein channel remains a significant challenge within the scientific community. Speculation surrounds its potential calcium ion selectivity, implicating a significant role in the pathology of COVID-19 by influencing viral replication and the cellular virus response.

ASC Dynamics Associated with ORF3a Protein Expression in Microvascular Lung Endothelial Cells

Proceedings of the Bulgarian Academy of Sciences
The NLRP3 inflammasome is a key sensor of cellular stress and pathogen-associated signals in the innate immune system. Using confocal fluorescence microscopy in microvascular lung endothelial cells, we examined ASC speck formation following expression of the SARS-CoV-2 ORF3a protein.
