Determination of Oxamniquine in Serum

Journal of Pharmaceutical Sciences, 1977
A method for the analysis of oxamniquine in serum (or plasma), sensitive to 10 ng/ml, was developed. Oxamniquine and a close structural analog as the internal standard were extracted from serum with ether. After derivatization with N,O-bis(trimethylsilyl)acetamide, oxamniquine was determined as its trimethylsilyl ether derivative by GLC using an ...

Synthesis and evaluation of new oxamniquine derivatives

International Journal of Pharmaceutics, 2002
Oxamniquine derivatives were synthesized and evaluated as novel schistosomicide agents. Oxamniquine (1,2,3,4-tetrahydro-2-[[(1-methylethyl)amino]methyl]-7-nitro-6-quinolinemethanol) was submitted to the Mannich reaction, using formaldehyde, paraformaldehyde and acetaldehyde as reagents, and gave three unexpected products: two of them were cyclized on ...

Fluorimetric determination of oxamniquine in biological fluids

Il Farmaco, 1999
A highly sensitive and specific fluorimetric method was developed for the determination of oxamniquine in biological fluids (urine and plasma). The proposed method is based on the reduction of oxamniquine using zinc/calcium chloride to obtain its nitroso derivative.

Seizures Associated with Oxamniquine Therapy

The American Journal of Tropical Medicine and Hygiene, 1986
Three patients who were treated for schistosomiasis mansoni with oxamniquine suffered generalized seizures. We suggest that this side effect may be more common than previously reported. Specific ethnic groups may be particularly at risk.

Tolerance of Kenyan Schistosoma mansoni to oxamniquine

Transactions of the Royal Society of Tropical Medicine and Hygiene, 1987
Although 30 mg/kg oxamniquine produced high levels (85.5 to 99.5%) of egg reduction in Kenyan children infected with Schistosoma mansoni after a single oral treatment, cure rates from children at Mwea in Kirinyaga district were lower than those from Machakos (58% v. 74%). Redosing uncured children confirmed this lower cure rate (36% v. 83%).

Studies on some derivatives of oxamniquine

Journal of Medicinal Chemistry, 1988
On the basis of the remarkable biological similarities between hycanthone and oxamniquine and as a sequel to our finding that some esters of hycanthone are active against hycanthone-resistant schistosomes, we prepared oxamniquine acetate, oxamniquine N-methylcarbamate, and four substituted phenylsulfonohydrazones of oxamniquine aldehyde.

Treatment of Complicated Schistosomiasis Mansoni with Oxamniquine

The American Journal of Tropical Medicine and Hygiene, 1978
Twenty male patients (mean age 23 years) with Schistosoma mansoni infections (mean egg count 429 +/- 311/g feces) were each treated with oxamniquine orally in a single daily dose of 20 mg/kg for 3 consecutive days. Seventeen patients had hepatosplenomegaly, two of these had ascites. Three patients had diffuse colonic polyposis, one of these had ascites.

The metabolism of oxamniquine—a new schistosomicide

Annals of Tropical Medicine & Parasitology, 1976
The metabolism of oxamniquine, 6-hydroxymethyl-7-nitro-2-isopropylaminomethyl-1,2,3,4-tetrahydroquinoline, has been studied in the mouse, rat, hamster, rabbit, rhesus monkey, dog and man. Urinary excretion is a major route of elimination in man. The compound is converted to two metabolites, the major one arising from oxidation of the 6-hydroxymethyl ...

The design of oxamniquine

2023
This chapter explores the development of oxamniquine by Pfizer pharmaceuticals, which the chapter uses as an example of how traditional strategies can be used in the development of a drug where the molecular target is unknown. It describes oxamniquine as an important drug in developing countries.

Chemotherapy of schistosomal colonic polyposis with oxamniquine

Transactions of the Royal Society of Tropical Medicine and Hygiene, 1978
Schistosomal polyposis of the colon is a common complication of schistosomiasis. In severe infections it may be associated with considerable morbidity. Surgery is associated with high morbidity and mortality. Niridazole therapy is of limited therapeutic value because of its hepatotoxicity but oxaminiquine has been found to be effective and safe ...
