Metformin Restores Mitochondrial Function and Neurogenesis in POLG Patient‐Derived Brain Organoids

open access: yesAdvanced Science, EarlyView.
Patient‐derived POLG‐mutant cortical organoids reveal neuronal subtype‐specific mitochondrial and synaptic defects, with dopaminergic neurons most affected. Metformin treatment restores neuronal identity, mitochondrial function, and excitability, increased mtDNA maintenance, and reprogrammed metabolism via TCA and redox pathways.

Pharmacophore Models and Pharmacophore-Based Virtual Screening: Concepts and Applications Exemplified on Hydroxysteroid Dehydrogenases

open access: yesMolecules, 2015
Computational methods are well-established tools in the drug discovery process and can be employed for a variety of tasks. Common applications include lead identification and scaffold hopping, as well as lead optimization by structure-activity ...

A rhodanine agent active against non-replicating intracellular Mycobacterium avium subspecies paratuberculosis. [PDF]

open access: yes, 2009
BACKGROUND: Antibiotic therapy targeting chronic mycobacterial disease is often ineffective due to problems with the emergence of drug resistance and non-replicating persistent intracellular antibiotic resistant phenotypes.

PHR‐Mediated Pi Starvation Response Mobile Messenger RNAs Represent Noncoding Transcripts in Recipient Tissues

open access: yesAdvanced Science, EarlyView.
Mobile messenger RNAs (mRNAs) can travel long distances, serving as systemic signals that participate in plant growth and stress acclimation. This work determines that PHR proteins mediate the Pi starvation response (PSR)‐specific long‐distance transport of mRNAs and that these PSR‐specific mobile mRNAs represent noncoding transcripts in recipient ...

4-Hydroxybenzoic Acid-Based Hydrazide–Hydrazones as Potent Growth Inhibition Agents of Laccase-Producing Phytopathogenic Fungi That Are Useful in the Protection of Oilseed Crops

open access: yesMolecules
The research on new compounds against plant pathogens is still socially and economically important. It results from the increasing resistance of pests to plant protection products and the need to maintain high yields of crops, particularly oilseed crops ...

The oxidoreductases POR and CYB5R1 catalyze lipid peroxidation to execute ferroptosis

open access: yesMolecular & Cellular Oncology, 2021
Our recent study showed that two oxidoreductases – NADPH-cytochrome P450 reductase (POR) and NADH-cytochrome b5 reductase (CYB5R1) – transfer electrons to oxygen to generate hydrogen peroxide (H2O2).

A multi-exon deletion within WWOX is associated with a 46,XY disorder of sex development [PDF]

open access: yes, 2011
Disorders of sex development (DSD) are congenital conditions where chromosomal, gonad or genital development is atypical. In a significant proportion of 46,XY DSD cases it is not possible to identify a causative mutation, making genetic counseling ...

The GMC superfamily of oxidoreductases revisited: analysis and evolution of fungal GMC oxidoreductases [PDF]

open access: yesBiotechnology for Biofuels, 2019
The glucose-methanol-choline (GMC) superfamily is a large and functionally diverse family of oxidoreductases that share a common structural fold. Fungal members of this superfamily that are characterised and relevant for lignocellulose degradation include aryl-alcohol oxidoreductase, alcohol oxidase, cellobiose dehydrogenase, glucose oxidase, glucose ...

Onion‐Mitochondria Inhibit Lipopolysaccharide‐Induced Acute Lung Injury by Shaping Lung Macrophage Mitochondrial Function

open access: yesAdvanced Science, EarlyView.
Proposed model of orally administered onion mitochondria (O‐Mit) uptake by lung macrophages and fuse with macrophage mitochondria (M‐Mit). This fusion reprograms the metabolism of dysfunctional M‐Mit in lipopolysaccharide‐induced murine acute lung injury by modulating dynamin‐related protein 1 (DRP1) phosphorylation and cardiolipin peroxidation ...

Genetic and Functional Evidence Links Germline Biallelic Inactivating Variants in WWOX to Histological Mixed‐Type Thyroid Cancer

open access: yesAdvanced Science, EarlyView.
The role of germline WWOX loss‐of‐function variants in oncogenesis has remained poorly characterized. This study provides the first comprehensive genetic and functional evidence that such variants promote tumorigenesis through the disruption of multiple tumor‐suppressive mechanisms.
