Reframing the Gut–Brain Axis: The Gut Wall as a Neural Immune Interface in Multiple Sclerosis and Autoimmune Neurological Diseases

open access: yesAnnals of Neurology, EarlyView.
Gut wall physiology in multiple sclerosis (MS) and autoimmune neurologic diseases remains underexplored but may span gut barrier dysfunction, enteric glial targeting, mucosal immune dysregulation, and autoimmune targets within the enteric nervous system (ENS).

Risk for Cardiovascular Adverse Events Associated With Sphingosine-1-Phosphate Receptor Modulators in Patients With Multiple Sclerosis: Insights From a Pooled Analysis of 15 Randomised Controlled Trials

open access: yesFrontiers in Immunology, 2021
BackgroundAll agents engaging sphongosine-1-phospate receptors (S1PRs) will have some cardiovascular effect. This study aimed to elucidate the risk of cardiovascular adverse events (AEs) in patients with multiple sclerosis (MS) treated with S1PR ...

Early‐stage health technology assessment of a curative gene therapy for multiple sclerosis

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aims Multiple sclerosis (MS) is associated with significant early morbidity, reduced life expectancy and substantial healthcare and societal costs. The primary objective of this study is to assess the early cost‐effectiveness potential of a novel gene therapy, IMMUTOL, for MS compared with current high‐efficacy treatment sequences.

Effect of the sphingosine-1-phosphate receptor modulator ozanimod on leukocyte subtypes in relapsing MS [PDF]

open access: yes, 2020
To better understand ozanimod's mechanism of action (MOA), we conducted exploratory analyses from a phase 1 study to characterize ozanimod's effect on circulating leukocyte subsets in patients with relapsing multiple sclerosis.

Harnessing Natural Killer Cells: From Neuroimmunology to Novel Therapies for Central Nervous System Diseases

open access: yesMedComm, Volume 7, Issue 10, October 2026.
This review delineates the multifaceted roles of natural killer (NK) cells in the central nervous system, spanning direct cytolysis, immunoregulation, and crosstalk with the brain microenvironment. We evaluate the translational potential and challenges of NK cell immunotherapies for a spectrum of neurological conditions.

IL‐7R‐Enriched Extracellular Vesicles From the Thymus Drive Colitis via Promoting Neutrophil Extracellular Trap Formation

open access: yesAdvanced Science, Volume 13, Issue 49, 3 September 2026.
Inflammatory bowel disease (IBD) pathogenesis remains unclear. This study reveals that thymus‐originated IL‐7R‐enriched extracellular vesicles exacerbate colitis by promoting neutrophil extracellular traps via a thymus‐gut axis, uncovering an inter‐organ communication mechanism in inflammatory bowel disease pathogenesis.

Ozanimod as a novel oral small molecule therapy for the treatment of Crohn's disease: The YELLOWSTONE clinical trial program

open access: yes, 2022
Background: Ozanimod, an oral sphingosine 1-phosphate receptor modulator currently approved for the treatment of moderately to severely active ulcerative colitis and relapsing multiple sclerosis, showed clinical, endoscopic, and histological benefit in ...

Comparative Effectiveness Analysis of Phase 3 Clinical Trials in Moderately to Severely Active Inflammatory Bowel Disease Reveals Increased Harm From Placebo and Challenges Equipoise

open access: yesJCC Plus, Volume 1, Issue 5, September 2026.
Across 47 Phase 3 trials in Crohn's disease and ulcerative colitis, placebo exposure was associated with measurable harm. Disease exacerbation events showed the greatest excess risk, with one additional exacerbation occurring for every 8–13 patients assigned to placebo rather than active therapy.

P037 Effect of Ozanimod Treatment and Discontinuation on Absolute Lymphocyte Count in Moderate-to-Severe Ulcerative Colitis: Results from a Phase 3 Trial

open access: yes, 2021
BACKGROUND: Ozanimod is an oral sphingosine 1-phosphate (S1P) receptor modulator selectively targeting S1P1 and S1P5, which reduces migration of lymphocytes involved in adaptive immunity from lymphoid tissues to blood and inflamed tissues while ...

Central Modulation of Selective Sphingosine-1-Phosphate Receptor 1 Ameliorates Experimental Multiple Sclerosis

open access: yesCells, 2020
Future treatments of multiple sclerosis (MS), a chronic autoimmune neurodegenerative disease of the central nervous system (CNS), aim for simultaneous early targeting of peripheral immune function and neuroinflammation.
