Elastase activity in experimental pancreatitis

The activity of elastase, elastase inhibitor and isozymes of lactate dehydrogenase (LDH) was studied in 11 dogs with experimental pancreatitis and 5 control dogs. All dogs with experimental pancreatitis died 6--14 hours after the induction of pancreatitis. Morbid anatomy studies proved severe pancreonecrosis.

Purification and characterization of ovine pancreatic elastase

Elastase was isolated from ovine pancreas and purified to homogeneity by two different procedures. One involved precipitation with ammonium sulphate, p-aminobenzamidine-Sepharose chromatography, CM-Sepharose ion exchange chromatography and S-300 Sephacryl chromatography.

Serum elastase I levels in pancreatic disease

The radioimmunoassay for human elastase I used in this study is accurate, sensitive, and specific, which we have confirmed. The assay can be done within 4 hours, which is important for clinical purposes. A total o 103 subjects were examined, and levels of 99 to 370 ng/dl (mean 200) in normal human sera were determined.

Inhibition of Pancreatic Elastase by Polyphenolic Compounds

Polyphenols are plant secondary metabolites commonly present in the human diet that possess the ability to bind and inhibit digestive proteins. In the present study, kinetic measurements of porcine pancreatic elastase (PPE) activity were determined using Suc-(Ala)(3)-p-nitroanilide as substrate and polyphenolic compounds as inhibitors.

Pancreatic elastase 1 is secreted from pancreatic acinar cells as one of the digestive enzymes. Pancreatic elastase 1 is present in serum in a complex formation with alpha 1-antitrypsin, and is measured by immunological methods. Serum pancreatic elastase 1 increases in acute pancreatitis and chronic relapsing pancreatitis.

Porcine and bovine elastins were digested by human leukocyte elastase and porcine pancreatic elastase. The enzymes showed similarities in the extent to which they digested elastin and the pattern and quantitative distribution of N-terminal amino acids in the digests.
