Filter Paper Chromatography

Consden, Gordon and Martin reported a very ingenious method for the separation of amino acids by filter paper chromatography in which phenol or some other appropriate organic solvent sweeps the applied amino acids along the filter paper, capillarity causing the organic solvent to move.

Paper Chromatography of Proteins [PDF]

Franklin and Quastel1 have recently applied the technique of paper chromatography to proteins. Protein–haemin complexes were prepared, and the position of each protein on the chromatogram could thus be revealed by the benzidine reaction. The method has limited application since many proteins do not react with haemin.

WE have recently developed a simple method for quantitative assay of amino-acids on the paper partition chromatogram. The method is in all probability applicable to any class of substance which can be readily located by means of a sensitive colourreaction on paper. Several technical points of general interest have arisen in the course of the work.
