Longitudinal circulating tumor DNA profiling in patients with advanced endometrial cancer using an off‐the‐shelf targeted NGS panel

open access: yesMolecular Oncology, EarlyView.
Intratumour heterogeneity complicates precision management of advanced endometrial cancer. Circulating tumor DNA (ctDNA) offers a minimally invasive strategy to capture tumor evolution and therapeutic resistance. Here, we compare tumor‐agnostic NGS with tumor‐informed ddPCR, outlining their relative sensitivity, concordance, and clinical implications ...

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

A Unified Parameter-Adaptive MPC Framework for Motion Control of Heterogeneous AGVs with Different Actuation Topologies

open access: yesActuators
The deployment of heterogeneous Automated Guided Vehicles (AGVs) in smart manufacturing requires control strategies that can accommodate distinct actuation characteristics and constraints.

Developmental programmes drive cellular plasticity, disease progression and therapy resistance in lung adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
This study shows that lung adenocarcinomas exploit developmental branching morphogenesis to acquire a therapy resistant basal‐like tumour cell state. This process was found to be regulated by combined TP53 loss‐of‐function and type‐I interferon signalling, identifying a novel axis for biomarker and therapeutic target discovery.

Lottery Rank-Pruning Adaptation Parameter Efficient Fine-Tuning

open access: yesMathematics
Recent studies on parameter-efficient fine-tuning (PEFT) have introduced effective and efficient methods for fine-tuning large language models (LLMs) on downstream tasks using fewer parameters than required by full fine-tuning.

Nonlocal Means Image Denoising With Minimum MSE-Based Decay Parameter Adaptation

open access: yesIEEE Access, 2019
Nonlocal means (NLM), a patch-based nonlocal recovery paradigm, has attracted much attention in recent decades. The decay parameter will greatly affect restoration performance of the NLM method.

Longitudinal genome‐wide aneuploidy measurements in circulating cell‐free DNA to predict lack of benefit from pembrolizumab in patients with metastatic urothelial cancer

open access: yesMolecular Oncology, EarlyView.
Many patients with urothelial cancer do not benefit from treatment with pembrolizumab, while at risk of severe side effects. Changes in the levels of circulating tumor DNA early during treatment, measured by a simple and affordable assay that can be easily implemented in the clinic, can be used as a prognostic tool to identify these patients.

Parameter-Efficient Adaptation of Qwen2.5 for Aspect-Based Sentiment Analysis Using Low-Rank Adaptation and Parameter-Efficient Fine-Tuning

open access: yesEngineering Proceedings
Aspect-based sentiment analysis (ABSA) plays a vital role in deriving fine-grained sentiment from textual content. As large language models (LLMs) are increasingly adopted for automated data annotation in natural language processing (NLP), concerns have ...
