Fabrication of solid lipid nanoparticles-based patches of paroxetine and their ex-vivo permeation behaviour

open access: yesArtificial Cells, Nanomedicine, and Biotechnology, 2023
Paroxetine is not suitable for oral administration due to its extensive first-pass metabolism, thus resulting in less bioavailability. This study aimed to prepare novel paroxetine-loaded solid lipid nanoparticles (SLNs) based sustained-release ...

Total synthesis of (±)-paroxetine by diastereoconvergent cobalt-catalysed arylation

open access: yes, 2014
A total synthesis of paroxetine is reported, with a diastereoselective and diastereoconvergent cobalt-catalysed sp3–sp2 coupling reaction involving a 3-substituted 4-bromo-N-Boc-piperidine (Boc = tert-butoxycarbonyl) substrate as a key step.

Opioid Toxicity Following Concomitant Use of Macrolide Antibiotics with Fentanyl, Hydromorphone, or Oxycodone: A Population‐Based Study

open access: yesClinical Pharmacology &Therapeutics, EarlyView.
Fentanyl, hydromorphone, and oxycodone are metabolized by cytochrome P450 3A4 (CYP3A4). Co‐administration with CYP3A4‐inhibiting macrolides (clarithromycin or erythromycin) may increase opioid concentrations and overdose risk, but the clinical impact of these interactions is unknown.
