Oxidative Stress in the Tumor Immune Microenvironment: Mechanisms and Therapeutic Perspectives
Oxidative stress is involved in several key processes in cancer, including redox regulation, DNA damage, post‐translational modifications, transcriptional regulation, epigenetic modifications, metabolic reprogramming, cell death, and immune modulation. These mechanisms collectively influence tumor progression, immune evasion, and therapeutic responses,

Molecular mechanism of poly(ADP-ribosyl)ation by PARP1 and identification of lysine residues as ADP-ribose acceptor sites [PDF]
Poly(ADP-ribose) polymerase 1 (PARP1) synthesizes poly(ADP-ribose) (PAR) using nicotinamide adenine dinucleotide (NAD) as a substrate. Despite intensive research on the cellular functions of PARP1, the molecular mechanism of PAR formation has not been ...

Targeting EZH2 in Cancer: From Molecular Mechanisms to Clinical Translation
The abnormal overexpression or gain‐of‐function mutations of EZH2 play a significant role in cancer occurrence and progression, highlighting the importance and potential of EZH2 as a cancer biomarker. Therefore, screening for effective and safe small‐molecule inhibitors, degraders, and natural compounds targeting EZH2 through preclinical cancer models ...

ABSTRACT We present the first x‐ray crystallographic structural evidence of an archaeal DNA ligase showing the AMP covalent adduct together with further cofactor hydrolysis, capturing a transient intermediary in the first step of the ligation reaction, triggered by the pyrophosphate hydrolysis.

Background and Purpose Endometriosis is a chronic, hormone‐dependent disorder characterized by ectopic implantation of endometrial tissue, often accompanied by pain and infertility. Although the progesterone receptor modulator RU486 is effective for pain relief, its impact on lesion regression is limited, possibly due to apoptosis resistance and ...

Intravesical delivery of p21 mRNA‐loaded lipid nanoparticles (p21‐LNP) restores the tumor suppressor p21 in bladder cancer. Localized p21 expression directly induces cell cycle arrest, DNA damage and apoptosis, and ultimately inhibits orthotopic tumor growth with minimal systemic distribution and favorable safety.

Update on PARP1 inhibitors in ovarian cancer
The clinical development of PARP inhibitors for the treatment of tumors deficient in BRCA1 or BRCA2 is based on the concept of synthetic lethality. From the initial proof of concept study with the PARP1 inhibitor olaparib (AZD2281) in BRCA mutation carriers, in which 28% of ovarian cancer patients achieved an objective response, the target population ...

Facile Synthesis of H‐L‐Photo‐Lysine and Its Genetic Incorporation Into Proteins
An efficient synthesis of a diazirine‐based noncanonical amino acid (ncAA), photo‐lysine (photoK), is reported, and its genetic incorporation into green fluorescent protein (GFP) is demonstrated. PhotoK‐enabled crosslinking is used to capture substrates and interaction partners of SIRT6 in vitro, in Escherichia coli, and in HEK293 cell lysates ...
